摘 要:目的:观察抗癌扶正方水提液与醇提液对荷Lewis肺癌小鼠的干预作用。方法:C57BL/6小鼠(雌雄各半)随机分为正常组(10只)、造模组(50只),将制得的Lewis肺癌细胞接种至C57BL/6小鼠右侧腋窝皮下,随机分为:模型组、顺铂组、水提组、醇提组,每组10只。顺铂组:隔日腹腔注射0.2mg/mL顺铂,0.2mL/只;水提组:灌服抗癌扶正方水提液(1g生药量/mL),0.2mL/只;醇提组:灌服抗癌扶正方水醇提液(1g生药量/mL),0.2mL/只;对照组与模型组灌服同等体积的生理盐水,连续给药24d。给药期间观察各组小鼠的一般反应,每4天称量一次体重。处死小鼠取血及脾脏,计算脾脏指数,检测白细胞含量,测量肿瘤体积和瘤重,观察肺组织病理变化情况。结果:抗癌扶正方不同提取液干预24天后,与模型组小鼠相比,水提组小鼠精神萎靡、反应迟钝等现象明显改善;肿瘤体积、瘤重及脾脏指数明显减小(P<0.05);白细胞明显升高(P<0.05);体重无明显变化;肺组织病理显示瘤细胞出现灶状坏死区及核碎裂象,并有炎性细胞浸润,瘤组织内及其周围微血管减少;与顺铂组相比,水提组小鼠的白细胞和脾脏指数都有显著提高(P<0.05),醇提组小鼠上述各项指标均无明显变化。结论:抗癌扶正方水提液对荷Lewis肺癌有抑制作用,醇提液则无抑制作用。Objective: To observe the invention effect of Fuzheng formula on Lewis lung carcinoma of mice. Methods: water extraction and ethanol extraction of Kangai C57BL/6 mice ( half male and half female) were randomly divided into 2 groups: a normal group (with 10 mice) and a model group (with 40 mice). Lewis lung carcinoma cells were inoculated to C57BL/6 mice on the right armpits subcutaneous. The mice were divided into 4 groups randomly: a model group, a cisplatin group, a water extraction group, and an ethanol extraction group, with 10 mice in each group. The cisplatin group: intraperitoneal injection of 0.2mg/mL cisplatin every other day, 0.2mL each. The water extraction, group: oral administrate water extraction of Kangai Fuzheng formula( lg/mL)0.2mL/only. Ethanol extraction group: administrated alcohol extraction of Kangai Fuzheng for- mula(lg/mL) orally 0.2mL each. The control group and the model group were fed with equal volume of saline for 24d successively. Observed the generally reaction of the mice of each group and weighed every four days dur- ing the administration period. After the mice were sacrificed, we took the blood and spleen, computed the spleen index, detect -ed the leukocytes, measured volume and weight tumor and observe pathological changes in lung tissue. Results: After 24 days intervention of water and ethanol extraction of Kangai Fuzheng formula, compared with the model group, the mice in the water extraction group were apathetic, unresponsive and im- proved significantly ; tumor volume, tumor weight and spleen index were significantly reduced (P〈0.05) ; white blood cells increased significantly (P〈0.05), but no significant changes in body weight. Lung tissue pathology showed tumor cells appear spotty necrosis and nuclear fragmentation, inflammatory cell infiltration and microvas- cular surrounding tissue reduced in tumor, compared with the cisplatin group, the spleen index and white blood cells increased significantly (P〈0.05).
