摘 要:目的:探讨贵州侗族健康人群MICB等位基因的分布特点。方法:收集100例健康无亲缘关系的贵州侗族人新鲜血液样本,采用世界卫生组织(WHO)推荐的标准盐析法从样本新鲜血液中提取基因组DNA,并用PCR-SSP、PCR-SBT两种方法对样本DNA进行MICB等位基因分型。结果:在贵州侗族人群中,检出7种MICB等位基因,其中MICB*005:02等位基因频率最高,其频率为58.50%,其次为MICB*002:01等位基因频率22.00%;而MICB*003及MICB*005:03等位基因频率最低,两者频率分别为1.50%。结论:贵州侗族人群MICB等位基因具有高度的多态性,该数据为研究MICB基因在同种异体器官移植和疾病易感性中的可能作用奠定了实验基础。Objective : To study the characterization of MICB alleles in a healthy Dong nationality of Guizhou. Meth-ods :Fresh blood samples were collected from one hundred health Dong population in Guizhou. Genomic DNA was ex-tracted from the peripheral blood using the standard salt precipitation method recommended by World Health Organiza-tion. MICB genotypes were determined by using PCR-sequence-specific primers (PCR-SSP) and PCR-sequence-based typing (PCR-SBT) , respectively. Results : Seven known MICB alleles in total were observed in the tested subjects. A-mong the characterized MICB alleles, MICB * 005 :02 showed the highest allele frequency (58. 5 0 % ) , followed by the allele frequency of MICB * 002 :01 (22. 00% ) . Furthermore, the allele frequency of MICB * 003 and MICB * 005 :03 were the lowest in the tested individuals ( 1.5 0 % ) . Conclusions : MICB allele showed high polymorphism in Dong na-tionality of Guizhou. These results could serve as the basis for future studies on the potential role of MICB in allogeneic organ transplantation and disease susceptibility in related ethnic groups.
