摘 要:目的:观察六味地黄丸干预阿霉素小鼠肾病对p38MAPK及NF-κB的蛋白表达的影响,探讨六味地黄丸防治阿霉素肾病的相关作用机制。方法:Balb/c小鼠27只,一次性尾静脉注射阿霉素10.5mg·kg^(-1)造模,实验分正常组7只、模型组10只、六味地黄丸组10只,六味地黄丸组给予六味地黄丸,正常组和模型组给予生理盐水,持续灌胃28d。生化检测方法观察小鼠24h尿总蛋白(UTP)含量以及血总蛋白(TP)、白蛋白(ALB)、尿素氮(BUN)、血肌酐(Scr)、胆固醇(CHOL)、甘油三酯(TG);光镜电镜观测肾组织形态变化;检测肾组织超氧化物歧化酶(Superoxyde dismutase,SOD)活性、丙二醛(Methane Dicarboxylic Aldehyde,MDA)含量;Western blotting观察小鼠肾组织p38MAPK及NF-κB的蛋白表达水平。结果:六味地黄丸干预组小鼠肾系数、24h尿总蛋白UTP、血TP、ALB升高;BUN、CHOL、TG均有显著降低(P<0.05);光镜和电镜显示肾脏病理改变改善;肾组织SOD活性明显增强,MDA含量明显下降;p-p38MAPK和NF-κB蛋白表达水平显著升高,p38MAPK表达没有变化(P<0.05)。结论:六味地黄丸防治阿霉素小鼠肾病的作用,可能与其可能抑制肾组织氧化应激,降低p-p38MAPK以及NF-κB p65蛋白表达水平相关。Objective:To observe the effect of Liuwei Dihuang Pill on the expression of p38MAPK and NF-κB in adriamycin-induced nephropathy,and to explore the mechanism of Liuwei Dihuang Pills in preventing and treating adriamycin nephropathy.Methods:27Balb/c mice were injected with doxorubicin 10.5mg·kg-1 at the end of the tail vein.The rats were divided into normal group(n=7),model group(n=10)and Liuwei Dihuang Pill(n=10).The intervention group was treated with Liuwei Dihuang Pill,the normal group and model group were given saline,continuously gavage 28 d.(TP),albumin(ALB),blood urea nitrogen(BUN),serum creatinine(Scr),cholesterol(CHOL),triglyceride(triglyceride)within 24 hours were measured by biochemical test.(SOD)activity,malondialdehyde(MDA)content in renal tissue were detected by Western blotting.The expression of p38MAPK and NF-κB in renal tissue of mice were detected by Western blotting.Results:The results showed that the levels of urinary protein UTP,blood TP and ALB were increased in the mice treated with Liuwei Dihuang Pill and the levels of BUN,CHOL and TG were significantly decreased(P〈0.05),and the renal pathological changes were improved by light and electron microscopy.The expression of p-p38MAPK and NF-κB protein was significantly increased and the expression of p38MAPK did not change(P〈0.05).Conclusion:Liuwei Dihuang Pill can prevent the oxidative stress and decrease the expression of p-p38MAPK and NF-κB p65 protein in renal tissue.
