摘 要:[目的]探讨白藜芦醇(Resv)对CXCL12诱导的人脐静脉细胞株(HUVEC)细胞增殖及血管生成的影响.[方法]取体外培养的HUVEC细胞,给予CXCL12和Resv干预后采用MTT法检测HUVEC细胞增殖情况,采用Matrigel胶体外管型形成实验检测HUVEC细胞管型形成情况,利用RT-PCR法检测HUVEC细胞血管内皮生长因子(VEGF)mRNA的表达水平.[结果]CXCL12可显著促进HUVEC细胞体外增殖、体外管型形成及VEGF mRNA表达(P<0.01,P<0.001);而Resv显著抑制CXCL12诱导的HUVEC细胞体外增殖、体外管型形成及VEGF mRNA表达(P<0.01,P<0.001).[结论]Resv可抑制CXCL12诱导的HUVEC细胞体外增殖及血管生成,其机制可能与下调VEGF mRNA表达水平有关系.OBJECTIVE To investigate the effects of resveratrol(Resv)on the proliferation and angiogenesis of human umbilical vein endothelial cells(HUVEC)induced by CXCL12.METHODS The HUVEC were cultured in vitro and MTT assay was used to detect the proliferation of the HUVEC intervened by CXCL12 and Resv.Matrigel plug assay was used to detect the tube formation of the HUVEC.RT-PCR was used to determine the levels of vascular endothelial growth factor(VEGF) mRNA.RESULTS CXCL12 could promote the proliferation and the tube formation of the HUVEC and up-regulate the mRNA expressions of VEGF in HUVEC(P〈0.01,P〈0.001).Resv significantly inhibited the effects induced by CXCL12(P〈0.01,P〈0.001).CONCLUSION Resveratrol could inhibit the proliferation and tube formation of HUVEC induced by CXCL12.The mechanism might be related with the down-regulation of VEGF mRNA expression.
