摘 要:目的对口岸截获的陈旧鼠样进行物种鉴定。方法运用DNA条形码技术对深圳盐田口岸入境,来自津巴布韦的集装箱内截获的腐败风干的鼠类残骸进行物种鉴定。分别从鼠眼周皮毛、腿部肌肉提取基因组DNA进行COI部分序列扩增,将扩增产物测序并比对,确定该样品物种。运用Mega 5.05软件构建系统发育树。结果经COI基因序列比对鉴定,截获鼠样为黑家鼠(Rattus rattus),与日本黑家鼠(GenBank号:AB752806)的Kimura 2-parameter遗传距离最近,值为0.003。结论 DNA条形码技术可以用于陈旧鼠样本的物种鉴定。Objective To identify the intercepted rat collected at frontier port. Methods Using DNA bareoding to i- dentify an intercepted rat which was eon'upt and dry in the container from Yantian port,Shenzhen city,came from Zimbabwe. Genome DNA was extracted from the fur around the eyes and the muselc tissues on the thigh,respective- ly. The COI gene was amplified by PCR and sequenced. The molecular phylogenetic tree was constructed by using Mega 5.05 software. Results The intercepted rat was Rattus rattus according to the molecular identification. The ge- netic distance between the sample and the black rat from Japan (GenBank accession number:AB752806) was the nearest,and the value was 0.003. Conclusion The DNA barcoding could be used to identify corrupted rat samples.
