摘 要:目的:比较高效液相色谱法(HPLC)、酶放大免疫法和微生物法测定血清万古霉素谷浓度的差异,确定三种测定方法间的线性关系。方法:分别采用HPLC、酶放大免疫法和微生物法对我院患儿血清中万古霉素谷浓度进行检测,比较分析测定结果,建立线性关系。结果:三种方法均能有效测定患儿万古霉素血药浓度,三种方法测得结果具有良好的相关性,相关性分析结果分别为:Y=1.006 9X+0.030 2(X-HPLC,Y-酶放大免疫法,r=0.999 9);Y=0.993 1X-0.043 8(X-酶放大免疫法,Y-微生物法,r=0.999 9);Y=1.000 1X-0.014 7(X-HPLC,Y-微生物法,r=0.999 9)。结论:HPLC、酶放大免疫法和微生物法均为可靠的万古霉素血药浓度检测方法,HPLC对于较低浓度的样本有更好的敏感性,各医疗机构可以根据条件选用。Objective: To explore the differences of plasma concentration of vancomycin by high performance liquid chromatography (HPLC), enzyme multiplied immune method and microbiological method. Methods: Microbiological method, enzyme immune method and HPLC were used to detect the plasma concentration of vancomycin, and clinical test results of these techniques were retrospectively analyzed. Results : There were no significant differences in the plasma concentration of vancomycin by microbiological method, enzyme immune method and HPLC. The outcomes of 3 methods showed good correlation and the regression equations were Y=1. 006,9X+ 0. 030,2, ( X-HPLC, Y-enzyme multiplied immune method, r = 0. 999,9 ) , Y= 0. 993,1X-0. 043,8, (X-enzyme multiplied immune method, Y-microbiological method, r = 0. 999,9 ) , Y= 1. 000,1X-0. 014,7, ( X-HPLC, Y- microbiological method, r= 0. 999,9 ). Conclusion: Microbiological method, enzyme multiplied immune method and HPLC are all effective and reliable for the plasma concentration detection of vancomycin. The hospitals can choose corresponding method according to their condition when determining plasma concentration of vancomycin.
