清肺利气通络法中药对哮喘模型大鼠肺组织白介素17表达的影响

摘　　要：目的探讨清肺利气通络法中药对卵清蛋白(OVA)致敏哮喘大鼠肺组织白介素17(IL-17)表达的影响。方法 40只雄性SD鼠随机分为空白组、模型组、布地奈德组和清肺利气通络颗粒组,每组10只。空白组不给予任何处理,其余3组动物均以OVA致敏和激发方案建立哮喘模型。模型组仅造模,不给予任何其他药物处理;布地奈德组在每次雾化激发前1h给予布地奈德混悬液雾化吸入15min;清肺利气通络组在每次雾化激发前分别给予清肺利气通络颗粒灌胃。末次激发4 h后处死大鼠,取肺组织进行病理切片,HE染色观察肺组织病理形态及免疫组化检测IL-17的表达。结果 HE染色镜下显示:与模型组比较,布地奈德组和清肺利气通络颗粒组的肺组织病理形态均有减轻。模型组、布地奈德组和清肺利气通络组大鼠肺组织中IL-17平均光密度(OD)值均较空白组显著增高(P<0.01);布地奈德组和清肺利气通络颗粒组大鼠肺组织中IL-17平均OD值较模型组显著降低(P<0.01);布地奈德组与清肺利气通络颗粒组大鼠肺组织中IL-17平均OD值比较差异无统计学意义(P>0.05)。结论以清肺利气通络法组方的清肺利气通络颗粒可以通过减轻哮喘大鼠肺组织IL-17的表达,改善肺组织病理形态,具有较好的效果,值得进一步深入研究。Objective： To study the effect of Qingfei Liqi Tongluo （QLT） method on the expression of interleukin - 17 in lung tissue of asthmatic rats induced by ovalbumin （OVA）. Methods ： 40 male SD rats were randomly divided into a blank group, a model group, a budesonide group and a QLT granule group, 10 rats per group. The blank group was not treated with any drug and the other three groups of the rats were induced by OVA to sensitization and stimulation to establish asthma models. The model group was only modeled with no other drug. The budesonide group was given inhalation of budesonide suspension for 15 min at 1 hour after each atomization. The QLT group was administrated with QLT granule before each atomization. The rats were sacrificed 4 hours later at the last stimulation, and their lung tissue was taken for pathological sections. The pathological changes of lung tissue were observed by HE staining and the expression of IL- 17 was detected by immunohistochemistry. Results： Compared with the model group, the histopathological changes of the lung tissue in the budesonide group The mean optical density （OD） of IL - 17 in the model group, higher than those in blank group （P 〈0.01 ）. But the mean OD group and QLT granule group was significantly lower than that in and QLT granule group decreased under HE staining. budesonide group and QLT group were significantly value of IL - 17 in the lung tissue of the budesonide the model group （P 〈0.01 ）. There was no statisti-cally significant difference （P 〉 0. 05 ） in the mean OD of IL - 17 in the lung tissue of the rats between the budesonide group and QLT granule group. Conclusion： QLT granule can ruduce the expression of IL - 17 in lung tissue of asthmat- ic rats and improve the pathological morphology of lung tissue, worthy of further study
