悬浮红细胞保存期末的质量检测

摘　　要：目的检测悬浮红细胞保存期末的质量情况。方法无菌接驳取悬浮红细胞标本,分别在保存前(0 d)和保存期末(35 d)进行红细胞压积(HCT)、血红蛋白(HGB)、红细胞计数(RBC)和游离血红蛋白检测,并使用透射电镜观察保存前和保存期末细胞形态结构的变化。结果在保存前(0 d)和保存期末(35 d)悬浮红细胞HCT、HGB、RBC等指标均符合质量要求,悬浮红细胞在保存前和保存期末HCT、HGB、RBC等指标变化不大,差异均无统计学意义(P>0.05);保存35 d后,红细胞游离血红蛋白浓度增高为(149.5±9.6)mg/L,与保存前的(26.5±1.9)mg/L比较,差异有统计学意义(P<0.05);透射电镜下观察其形态无明显变化,红细胞为双凹圆盘状,细胞膜结构清楚,膜表面光滑完整,未见突起、凹陷、空泡、缺失等膜损伤。结论悬浮红细胞保存前和保存期末各项检测指标均符合质量要求;控制悬浮红细胞制备和保存中各个环节,对于保护红细胞的形态和功能,保证血液质量,保证血液输注效果,均具有重要意义。Objective The aim of this study was to investigate the quality of the suspension red blood cells（SRBCs） whichwere stored for 35 days. Methods The storage cell characteristics were examined on days 0 and 35. We measured thebiochemical parameters including hematocrit（HCT）,hemoglobin（HGB）,red blood cell（RBC）and free hemoglobin（FHb）0 We also observed the structural changes of the red blood cells by using transmission electron microscope. Results Theresults showed that HCT,HGB and RBC levels of suspended red blood cells did not significantly change after 35 daysstorage（P0.05）. The values of free hemoglobin（FHb）concentrations increased,and the consequences were staticallysignificant（P0.05）. The structure of the red blood cells was normal. Red blood cells were double concave disc. Themembrane structure was clear,while the membrane surface was smooth and complete,and the membrane damage such asbumps,lack of the sag or cavitation was not found. Conclusion SRBCs were qualified for clinic transfusion after storagefor 35 days. It is important to control the conditions of preparation and storage of suspension red blood cells for the quality ofblood and the effect of blood transfusion.
