摘 要:目的:探讨甲基莲心碱对紫外线致人成纤维细胞形态结构的影响,为防治皮肤光老化性疾病提供实验依据。方法:建立中长波紫外线辐射人皮肤成纤维细胞光老化模型,实验分为对照组、模型组(单纯紫外线照射)和药物组(紫外线照射加甲基莲心碱干预),观察和分析成纤维细胞经紫外线照射后细胞生长形态和超微结构变化。结果:紫外线照射成纤维细胞后,细胞失去正常形态,超微结构显示线粒体损伤,而经甲基莲心碱处理后细胞损伤明显减轻。结论:适当浓度(0.8μM)的甲基莲心碱能减轻紫外线照射后成纤维细胞的结构损伤,为防治皮肤光老化疾病提供了新的治疗思路。Objective: This study discussed the effect of Neferine on the ultrastructure of ultraviolet to fibroblasts, and provided experimental basis for skin photoaging treatment.Methods: Human skin fibroblasts photoaging model was established by UVA and UVB radiation.Fibroblasts divided into normal control group, model group(radiated with UVA and UVB only) and treatment group(radiated with UVA and UVB and treated with Nef).Morphological and ultrastructural alterations of fibroblasts following UV irradiation were observed and the impact of Nef on it was explored.Results: Fibroblasts following UV exposure lose its normal shape and ultrastructural analysis also reveals damage to mitochondria;nevertheless, the damage was reduced when treated with Nef.Conclusions:The appropriate concentration of neferine can reduce damage of the ultrastructure of fibroblasts after ultraviolet irradiation, and provide a new method for the prevention and treatment of skin photoaging diseases.
