双醋瑞因对大白兔肥厚黄韧带天狼猩红检测结果的影响研究

摘　　要：目的观察双醋瑞因对黄韧带肥厚大白兔天狼猩红检测结果的相关影响。方法把黄韧带肥厚造模成功的动物分成A与B两组,A组是模型组,B组是药物组,A组应用蒸馏水进行灌胃,B组应用双醋瑞因进行灌胃,剂量是3.8mg/kg,连续应用时间为3个月。两组在第4周、第8周、第12周对局部的黄韧带实施病理切片天狼星红染色,使用偏振光显微镜,对其胶原纤维的变化进行观察。并在Qwin V3图像分析软件中统计胶原面积。结果偏振光观察天狼猩红染色显示,正常的黄韧带组成以弹性纤维为主,弹性纤维与胶原纤维的比例大概为2:1,胶原纤维主要以Ⅲ型胶原为主,并且有少量的Ⅰ型胶原。在模型组中,胶原纤维的比例会渐渐地增加,在第4周,Ⅰ型胶原和Ⅲ型胶原是比较相似的,在第8周的时候,Ⅰ型胶原的比例显著地升高了,Ⅲ型胶原渐渐地降低,在第12周Ⅲ型胶原出现了大量的增生情况,Ⅰ型胶原的比例显著地减少。在药物组,第4周的时候Ⅰ型胶原和Ⅲ型胶原要比正常组的黄韧带有所升高,在第8周与第12周的阶段,Ⅰ型胶原的比例显著地增加,特别是在第12周的时候,但是和模型组进行对比,增加显著的得到了抑制。结论对于肥厚的黄韧带中胶原纤维增生双醋瑞因能够起到较强的抑制作用,从而对较好地改善了肥厚的黄韧带中的组织结构,让其恢复到正常水平或是接近正常的状态。Objective To investigate the effect of diacerein on rabbit yellow ligament hypertrophic sirius red detection.Methods The yellow ligament hypertrophy model animals was maded successful,which were divided into two groups, group A is the model group, group B is the drug group.Group A： pour distilled water into the stomach, Group B ： pour diacerein into the stomach, the dose was 3.Smg/kg, each group was intervened by three months.local yellow ligament was detected in the 4 weeks, 8 weeks and 12 weeks by histology with Sirius red staining, then it was observed under polarized light microscopy, use the QwinV3 image analysis software for statistical.Results In the normal yellow ligament, Elastic fibers and collagen fiber ratio is about 2：1, Collagen fibers were mainly collagen type III, and there was a small amount of collagen type I.In model group, the percentage of collagen fibers gradually increased, in 4 weeks, collagen m is almost equal to collagen I , on 8 weeks, the proportion of collagen I was increased significantly, while collagen III gradually reduced, on the twelfth week , the collagen El was increased significantly, while collagen El gradually reduced.In the drug group, on 4weeks, collagen was compared collagen I were almost equal to normal group, on 8 and 12 weeks, collagen I increased significantly, especially on 12 weeks however, compared with the model group, the increase was significantly inhibited.Conclusion Diacercin can inhibit collagen fiber hyperplasia in hypertrophic ligamentum flavum, so as to improve the structure of hypertrophic liganaentum flavum and restore it to normal or near normal state..
