摘 要:2014年-2015年陕西杨凌地区部分商品蛋鸡场产蛋量下降了10%~40%,剖检部分鸡只发现肝脏肿大出血。为确诊该疫情的病因,从发病的6个蛋鸡场采集了337份血清和268份粪便,利用间接ELISA和套式RT-PCR分别对血清中的禽戊型肝炎病毒(HEV)抗体和粪便中的禽HEV RNA进行了实验室检测。ELISA检测结果发现,168份血清为禽HEV抗体阳性,阳性率为49.85%;套式RT-PCR检测结果发现,63份粪便为禽HEV RNA基因片段阳性,阳性率为23.51%。基因片段的序列同源性分析发现,杨凌地区的禽HEV分离株与国内的同源性为93.0%~99.2%;进化树分析表明,其与国内其他地区的分离株在同一进化分支上,同属禽HEV基因3型。In 2014-2015, many laying hen flocks in Yangling of Shaanxi province had a decrease in egg production (10%-40%) in laying hens.Some necropsied laying hens showed liver hemorrhage and enlarged.To determine the causative agent of the disease, the 337 serum and 268 fecal samples were collected from 6 laying hen flocks.Subsequently,anti-avian hepatitis E virus (HEV) IgG antibodies in serum samples by in- direct ELSIA and avian HEV RNA by nested RT-PCR (RT-nPCR) were tested. The results of indirect ELISA showed that the 168 serum samples were positive for anti-avian HEV IgG antibodies and the positive rate was 49.85%. The results of RT-nPCR showed that the 63 fecal samples were positive for avian HEV RNA and the total positive rate was 23.51%.Comparisons of the sequences from the positive fecal samples showed that they shared 93.0%-99.2% identities with the isolates from other provinces of China. Phylogenetic analysis showed that these strains were located in the same branch with other isolates in China and they all belonged to genotype 3 avian HEV.
