摘 要:目的:观察三叶青黄酮诱导肺癌SPC-A-1细胞凋亡作用和对cleaved-caspase-3表达的影响,探讨其抗肺癌的可能作用机制。方法:体外培养SPC-A-1细胞,以不同剂量三叶青黄酮进行处理,未给予三叶青黄酮组作为对照组。采用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)比色分析法,观察三叶青黄酮对SPC-A-1细胞增殖抑制作用,计算半数抑制浓度(IC50);采用流式细胞仪和TUNEL染色检测三叶青黄酮对SPC-A-1细胞凋亡的影响;分别采用免疫细胞分析和蛋白印记分析(Western blot)检测三叶青黄酮对SPC-A-1细胞cleaved-caspase-3表达的影响。结果:三叶青黄酮对SPC-A-1细胞增殖具有明显抑制作用,呈剂量和时间依赖性,并诱导SPC-A-1细胞凋亡,与对照组比较,差异具有统计学意义(P<0.05或P<0.01);三叶青黄酮显著增加SPC-A-1细胞cleaved-caspase-3表达水平,与对照组比较,差异具有统计学意义(P<0.05或P<0.01)。结论:三叶青黄酮可显著抑制SPC-A-1细胞增殖,并诱导其凋亡,机制可能与增强cleaved-caspase-3表达有关。OBJECFIVE To observethe effects of radix tetrastigma hemsleyani flavones on induced apoptosis and expression of cleaved-caspase-3 in lung cancer SPC-A-1 cells, and discuss its possible mechanisms of anti-lung cancer. METItOE6 SPC-A- I cells were cultured in vitro and treated with radix tetrastigma hemsleyani flavones at various concentrations. Control group without radix tetrastigma hemsleyani flavones was also assigned. The proliferation of SPC-A-1 cells and the 50% inhibitory con- centration (IC50) were measured by 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) colorimetric assay. The apoptosis of SPC-A-1 cells was observed by flow cytometry and TUNEL staining. The expression of cleaved-caspase- 3 was detected by immunocytochemical analysis or western blot analysis. RESULTS Compared with control group, radix tet- rastigma hemsleyani flavones inhibited significantly the proliferation of SPC-A-1 cells and induced their apotosis in a dose and time dependent manner (P〈0. 05 or P〈0. 01), and also increased significantly the expression and cleaved-caspase-3 in SPC-A- 1 cells (P〈0. 05 or P〈0. 01). CONCLUSION Radix tetrastigrna hemsleyani flavones can inhibit the proliferation of SPC-A-1 cells and induce their apotosis, and its possible mechanisms may be related to enhanced the expression of cleaved-caspase-3.
