摘  要:目的探讨健康人和2型糖尿病(T2DM)患者血清游离脂肪酸(FFA)之间的相关关系,确定潜在生物标志物。方法采用气相-质谱联用(GC-MS)方法检测56例健康体检者(对照组)和57例2型糖尿病患者(2型糖尿病组)血清游离脂肪酸水平,结合PCA和PLS-DA方法对检测出的游离脂肪酸进行分析。结果 2型糖尿病组高密度脂蛋白(HDL-C)水平较对照组显著降低(P<0.05),葡萄糖(GLU)、血清甘油三酯(TG)、低密度脂蛋白(LDL-C)胆固醇(CHOL)水平显著升高(P<0.01);十二烷酸(C12:0)、棕榈酸(C16:0)、反-油酸(C18:1T)、亚油酸(C18:2)、花生酸(C20:0)、二十碳五烯酸(C20:5)、γ-亚麻酸(γ-C18:3)水平较对照组显著降低(P<0.05),硬脂酸(C18:0)、花生一烯酸(C20:1)、二十碳二烯酸(C20:2)、二十碳三烯酸(C20:3n-6)、花生四烯酸(C20:4)、山嵛酸(C22:0)、二十二碳五烯酸(C22:5n-3)、二十二碳六烯酸(C22:6ns)水平较对照组显著降低(P<0.01),其他各游离脂肪酸无显著性差异(P>0.05)。结论 2型糖尿病的发生与体内脂代谢紊乱有关,以C18:1、C18:2和C16:0作为2型糖尿病潜在生物标志物。Objective To explore the correlation between serum free fatty acids in healthy people and type 2 diabetic patients, and to identify potential biomarkers. Methods Used gas chromatography/mass spectrometry ( GC - MS) method for detecting serum free fatty acid levels from 56 healthy subjects ( control group) and 57 cases of patients with type 2 diabetes ( type 2 diabetes group). PCA and PLS - DA methods were performed to do statistical analysis. Results High - density lipoprotein ( HDL - C) level of type 2 diabetes group was significantly lower than that of the control group ( P 〈 0.05 ), and glucose ( GLU), serum triglyceri - de ( TG), low - density lipoprotein ( LDL - C ), cholesterol (CHOL) levels were significantly increased (P 〈 0.01 ). Aurie acid ( C12:0), palmitic acid ( C16:0), oleic acid ( C18 :IT), linoleic acid ( C18:2 ), arachidonic acid ( C20:0), eicosapentaenoic acid ( C20 : 5 ) , -/- linolenic acid ( ",/ - C18 : 3 ) were significantly lower than that of the control group ( P 〈 0. 05 ). The levels of ste - aric acid ( C18 : 0 ), peanut acid ( C20 : 1 ), eicosadienoic acid ( C20: 2 ), eicosatrienoic acid ( C20 : 3n - 6 ), arachidonic ( C20 : 4 ), docosanoic acid ( C22 : 0 ), timnodonic acid ( C22 : 5n - 3 ) , docosahexenoic acid (C22:6ns) were significantly decreased (P 〈 0.01 ). No significant difference was observed in other free fatty acids ( P 〉 0.05). Conclusion The occurrence of type 2 diabetes is related to the disorder of lipid metabolism, and C18 : 1, C18:2 and C16:0 can be used as potential biomarkers for type 2 diabetes.
