益气健脾祛痰化瘀法对脾虚痰浊动脉硬化巴马猪水液代谢的影响  被引量:3

摘  要:目的:观察脾虚痰浊动脉硬化(AS)巴马小型猪水液代谢的变化及益气健脾、祛痰化瘀法的作用。方法:将20头巴马小型猪随机分组,正常组、模型组、益气健脾组及益气健脾祛痰化瘀组各5头。以高脂饮食、跑步劳倦结合冠脉球囊拉伤法制备脾虚痰浊AS病证结合模型至24周。正常组巴马小型猪喂正常饲料,模型组巴马小猪行冠脉球囊拉伤后予高脂饲料结合跑步过劳制备脾虚痰浊AS模型。益气健脾组及益气健脾、祛痰化瘀组在模型组制备基础上,于高脂饲料中添加相应的中药颗粒剂。观察48周后各组巴马小猪的血清钾、钠、氯、尿素氮、肌酐的含量,血清水液代谢调节激素精氨酸加压素(AVP)、心房利钠肽(ANP)、醛固酮(ALD)以及肾组织水通道蛋白(AQP)1、2、3、4、钠钾ATP酶蛋白(Na^+-K^+-ATPase)表达变化。结果:与正常组相比,模型组血清钾、钠、氯、尿素氮、肌酐水平无变化(P>0.05),ANP、AVP上调(P<0.05),ALD有上调趋势(P>0.05),模型组肾组织AQP1、2、3及钠钾ATP酶蛋白表达下调(P<0.05),AQP4表达上调。与模型组比较,益气健脾、祛痰化瘀组ANP下调(P<0.05),AVP、ALD下调(P>0.05),益气健脾组AVP下调(P>0.05);益气健脾组及益气健脾、祛痰化瘀组肾组织AQP1、2、3表达均上调(P<0.05),AQP4表达下调;益气健脾组钠钾ATP酶蛋白表达上调(P<0.05)。结论:脾虚痰浊AS模型未见血清离子紊乱,但水液代谢调节激素中ANP、AVP、ALD有上调,肾组织AQP1、2、3下调,下调,AQP4表达上调;益气健脾及益气健脾祛痰化瘀法可上调肾组织AQP1、2、3,下调AQP4。益气健脾祛痰化瘀法可下调水液代谢调节激素,益气健脾法可上调钠钾ATP酶。益气健脾、祛痰化瘀法在恢复水液代谢调节激素方面优于益气健脾组。Objective:To observe the changes of water metabolism and effect of the benefitting spleen Qi and elimina- ting phlegm and blood stagnant treatment to bana minipig with spleen dificiency and phlegm type of atherosclemsis. Meth- od: Twenty bama minipigs were randomly divided into normal group, model group ,jianpiyiqi group and yiqijianpiqutan- huayu group with five in each. The spleen dificiency and phlegm atheroselerosis model were instant by high fat diet com- bined tied by running after balloon injury in the coronary artery until 24 weeks, the bama minipigs of normal group were fed normal basic diet, the bama minipigs of yiqijianpi group and yiqijianpiqutanhuayu group were add Chinese herb on the basis of normal fodder of the model group for 48 weeks. Observe K^+ , Na ^+ , CL ^- , Bun, Cr, ALD, AVP and ANP in blood and the protein expression of aquaporins (AQP) 1,2,3,4 and Na ^+ - K ^+ - ATPase in the mini - pigs kidney after 48weeks. Result : Compared with normal group, content of K ^+ , Na^+ , CL ^-, Bun, Cr in blood cannot Show the obvious differ- ent(P 〉0.05) ,ANP,AVP,ALD of model group were higher than the normal group,but only ANP and AVP show the obvious different( P 〈 0.05 ) , ALD have not obvious different o expression of AQP1,2,3 and Na + - K + - ATPase reduced ( P 〈 0. 05 ), expression of AQP4 increased( P 〈 0. 05 ). Compared with model group, yiqijianpi and yiqijianpiqutanhuayu group increased expression of AQP1,2,3 in the bama mini - pigs kidney( P 〈 0.05 ). and yiqijianpi group increased Na ^+ - K ^+ - ATPase ( P 〈 0. 05 ) , yiqijianpiqutanhuayu group reduced ANP of blood. Conclusion : Spleen dificiency and phlegm atherosclerosis model show ANP,AVP of blood higher than normal group, and AQP1,2,3, Na ^+ - K ^+ - ATPase reduced than normal group, AQP4 increased than normal group.
