重楼总皂苷对A549细胞凋亡及Caspase3、Bcl-2蛋白表达的影响

摘　　要：目的:探讨重楼总皂苷诱导人肺腺癌A549细胞凋亡的作用及对Caspase3、Bcl-2蛋白表达的影响。方法:CCK-8法检测重楼总皂苷对细胞增殖的抑制作用;Hoechst染色后激光共聚焦显微镜观察细胞凋亡形态的变化;Annexin V-FITC/PI双标记流式细胞术检测重楼总皂苷对细胞凋亡的影响;Western blotting检测重楼总皂苷对细胞凋亡相关蛋白Caspase3、Bcl-2表达的影响。结果:重楼总皂苷对A549细胞的生长有显著抑制作用,可引起细胞形态发生明显改变,诱导细胞发生凋亡,显著上调促凋亡蛋白Caspase3的表达,下调抑制凋亡蛋白Bcl-2的表达,且与浓度成依赖关系。结论:重楼总皂苷可显著诱导A549细胞凋亡,其机制与上调促凋亡蛋白Caspase3的表达,下调抑制凋亡蛋白Bcl-2的表达相关。Objective： To investigate the Rhizoma Paridis total saponins on apoptosis of human lung adenocarcinoma A549 ceils and its effect on the expressions of Bcl - 2 and Caspase3. Methods ： Detect Rhizoma Paridis total saponins on （：ell proliferation inhibition by CCK -8. After Hoechst staining, laser confocal microscope was used to observe cell mor- phological changes. Detect the influence of the total saponins from Rhizoma Paridis on cell apoptosis by annexin V/PI double labeled flow cytometry. Detect Rhizoma Paridis total saponins on apoptosis related Caspase3 and bcl -2 expres- sions by Western blotting. Results： Rhizoma Paridis total saponins on A549 cell growth has a significant inhibitory effect and can cause the cell morphology changed significantly, induce apoptosis, significantly up - regulate promoting Caspase3 expressions and inhibit the expression of Bcl -2 protein and and concentration into dependencies. Conclusion： Rhizoma Paridis total saponins can significantly induce the apoptosis of A549 cells and its mechanism may be related to the up reg- ulation of Pro Caspase3 expression and downregulating the expression of Bcl -2 protein.
