摘 要:目的:探讨济生肾气丸对高尿酸血症大鼠的影响及作用机制。方法:Wistar雄性大鼠90只,随机分为6组:正常组、模型组、济生肾气丸高剂量组、中剂量组、低剂量组、苯溴马隆组。除正常组外,其他5组Wistar雄性大鼠均应用氧嗪酸钾灌胃法制备高尿酸血症模型。造模开始后14 d,按组别给药。于灌胃后14、28、56 d取眼眶血,于56 d处死小鼠后进行肾脏取材。测定大鼠血尿酸水平,应用Western-Blot法测定小鼠肾脏URAT1和OAT1蛋白表达含量。结果:(1)治疗时间长短、不同药物及剂量对大鼠血尿酸水平均有影响,济生肾气丸各剂量组大鼠血尿酸水平下降,明显低于造模组(P<0.05),且中剂量(300 mg/kg)济生肾气丸对血尿酸影响最大。(2)济生肾气丸中剂量组(300 mg/kg)和苯溴马隆组大鼠URAT1的蛋白表达均降低(P<0.05),OAT1的蛋白表达均升高(P<0.05),且济生肾气丸中剂量组(300 mg/kg)效果较佳。结论:济生肾气丸可降低高尿酸血症大鼠血尿酸水平,其中济生肾气丸中剂量(300 mg/kg)效果最佳,其作用机制可能是降低URAT1的蛋白表达、升高OAT1的蛋白表达。Objective : This study aims to observe the effect of Jisheng Shenqi Pills on the treatment of rats with hyperu- ricemia in blood uric acid and explore its mechanism. Method: Ninety male Wistar rats were randomly divided into 6 groups : normal group, model group, Jinsheng Shenqi pill high - dose group, middle - dose group, low - dose group and benzbromethale group. In addition to the normal group, rats in the other five groups were treated with potassium oxonate potassium by garage to prepare the hyperuricemia model. 14 days after the start of modeling, rats were fed with drugs ac- cording to group administration. Blood was collected from eyes at the end of 14 days, 28 days and 56 days. Rats were ex- ecuted at the end of 56 d. The levels of blood uric acid was measured and the expressions of URAT1 and OAT1 protein in kidney were measured by Western - Blot method. Results:①The period of treatment can affect the level of blood uric acid. Both drugs and doses have an impact on blood uric acid levels(P 〈0. 05). The middle- dose group had the best effect on decreasing blood uric acid, and it was significantly lower than that of the model group (P 〈 0. 05 ). ②The pro- tein expressions of URAT1 in the middle dose group (300 mg/kg) and the bromocriptine group were significantly lower than those of the other control groups ( P 〈 0.05 ). The protein expression of OAT1 in two groups were significantly higher than those of the other control groups ( P 〈 0. 05 ). And the middle - dose group had the best effect. Conclusion : Jisheng Shenqi Pills can reduce the level of blood uric acid in rats with hyperuricemia. Middle - dose of Jisheng Shenqi Pills (300 mg/kg) has the best effect. The mechanism of Jisheng Shenqi Pill to reduce blood uric acid level may be relying on reducing the expression of URAT1 protein and increasing the expression of OAT1 protein.
