摘  要:目的:通过提取纯化得到太子参精制多糖,并对其单糖组成进行鉴定分析。方法:水提醇沉后的多糖采用Sevag法除去蛋白,再经Sephadex G-100凝胶柱层析进一步纯化,透析冻干后制得纯度较高的精制多糖。高温强酸条件下将多糖进行水解,加入1-苯基-3-甲基-5-吡唑啉酮(PMP)溶液衍生化后采用HPLC法分析单糖组成。结果:精制太子参多糖的含量为82.30%,主要由葡萄糖、半乳糖、木糖、鼠李糖等单糖组成。结论:本研究可为太子参药材质量标准的提升完善提供基础。Objective: To obtain high purity polysaccharides from radix pseudostellariae by extraction and purification process,then analyze the monosaccharide composition of the polysaccharides. Methods: The crude polysaccharide was prepared by water-extraction and alcohol-precipitation method,then proteins was eliminated by Sevag method. Crude sample was further purified by using Sephadex G-100 gel column chromatography. High purity polysaccharides powder was finally obtained by casting the dialysed solution into freeze dryer. The polysaccharide was hydrolyzed to monosaccharide in high-temperature and acidic condition,then the hydrolysate was derivatized with 1-phenyl-3-methyl-5-pyrazolone( PMP) and analyzed by HPLC. Results: The content of high purity polysaccharides powder is 82.30%. It mainly comprises glucose,galactose,xylose and rhamnose. Conclusion: This research can provide basic reference information for the improvement of follow-up quality standard of radix pseudostellariae.
