摘 要:目的:从蛋白质组学的角度探讨C-反应蛋白(CRP)损伤内皮细胞(EC)的作用机制。方法:采用培养的人冠状动脉内皮细胞HCAEC第4代用于实验,25μg/mL CRP刺激HCAEC 24 h,2D DIGE结合MALDI-ToF-ToF质谱鉴定进行蛋白质组学分析研究。结果:发现差异表达蛋白斑点11个,鉴定出其中4个,包括Heat shock cognate 71 kDa protein、Elongation factor 1-beta和Proteasome activator complex subunit 1等。结论:通过蛋白质组学研究,筛选了CRP损伤EC的差异表达蛋白,为深入理解EC损伤的分子机制提供有益的线索。Objective: To investigate the role of CRP in endothelial cell by proteomics analysis. Method: Human coronary artery endothelial cells were used at passage 4.25 μ g/mL CRP served as stimulus for endothelial cell injury.Differentially expressed proteins were analyzed by 2D DIGE and identified by MALDI- ToF-ToF MS analysis.Result: Differentially expressed 11 spots were visualized and 4 were identified, including Heat shock cognate 71 kDa protein, Elongation factor 1-beta and Proteasome activator complex subunit 1.Conclusion: This study screen differentially expressed proteins by CRP stimulus via proteomics analysis, providing useful understand of the underlying molecular mechanism of endothelial cell injury.
