糙皮桦‘Jacquemontii’茎段离体快速微繁殖技术研究

摘  要:以糙皮桦‘Jacquemontii’带腋芽茎段为外植体,以WPM为基础培养基,添加不同类型及质量浓度的植物生长调节剂,进行诱导茎段的腋芽萌发、丛芽增殖及生根等一系列组织培养研究,以筛选出适合‘Jacquemontii’组织培养的最优培养基配方,成功获得健壮的再生植株。结果表明,腋芽茎段最佳诱导培养基:WPM+1.5 mg/L 6-BA+2.5mg/L GA3,其萌芽率达71.67%;最佳增殖培养基:WPM+3.0 mg/L 2-ip+0.5 mg/L IAA+1.5 mg/L GA3,其增殖倍数为8.12;最佳生根培养配方:1/2WPM+0.8 mg/L IBA+0.5 mg/L NAA+0.5 mg/L活性炭,生根数为3—6条,生根率达97.92%。On the basis of WPM,the axillary buds of stem segments of Betula utilis var. ‘Jacquemontii'were used as explants to culture by adding different types and concentration ratio of plant growth regulator to induce a series of processes of germination of axillary buds and proliferation,rooting and so on,to select the optimal culture medium formula which is suitable for ‘Jacquemontii',successfully getting robust generated plantlet. The results show that the best initiation medium for the axillary buds was WPM+1. 5 mg/L BA + 1. 0 mg/L GA3,the induction rate of which is 71. 67%. The optimum proliferation medium was WPM + 3. 0 mg/L 2-ip + 0. 5 mg/L IAA + 1. 5 mg/L GA3,the proliferation multiple of which was8. 12. The best rooting medium was 1/2 WPM + 0. 8 mg/L IBA + 0. 5 mg/L NAA,root number is about 3-6,rooting rate reached 97. 92 %.
