摘 要:目的:探讨氧化低密度脂蛋白(ox-LDL)对人脐静脉内皮细胞(HUVEC)孤儿核受体NOR1表达的影响。方法:采用ox-LDL建立人脐静脉内皮细胞损伤模型,将细胞分为空白组、ox-LDL高、中、低剂量组(25μg/ml、50μml、100μg/ml、200μg/ml),采用CCK8法检测HUVEC活性,采用实时荧光定量PCR分析NOR1转录水平变化,采用Western-Blot分析NOR1蛋白表达的变化。结果:在HUVEC中加入不同浓度ox-LDL共同培养12h后,CCK8值与ox-LDL呈现明显的剂量反应关系,与空白对照组相比具有显著性差异(P<0.01)。Ox-LDL能够诱导NOR1 mRNA转录和蛋白表达,NOR1 mRNA峰值出现在ox-LDL共同培养后的3h,NOR1蛋白质最大值出现在ox-LDL共同培养后的6 h。结论:ox-LDL能够损伤HUVEC,并能诱导NOR1的表达上调,其在内皮细胞损伤和动脉粥样硬化炎性反应中的作用有待进一步研究。Objective The goal of the present study was to determine the effects of oxidized low density protein( ox-LDL)upon the NOR1 expression. Method A model of ox-LDL-induced injury human umbilical vein endothelial cell was established. The cells were co-incubarated with four concentration of ox-LDL( 25,50,100 and 200 μg/ml). The viability of cell were determined by CCK8 method. The NOR1 mRNA and protein were measured by Using QRT-PCR and western blotting techniques. Results Our data indicated that after co-incubarated with ox-LDL for 12 h,the viability,NOR1 expression of Human umbilical vein endothelial cell was reduced by ox-LDL in a dose-dependent manner. Rapid temporal changes in NOR1 mRNA and protein levels were observed following ox-LDL treatment with levels peaking at 3 h and 6 h respectively. Conclusion Ox-LDL induced injury of human umbilical vein endothelial cell and increased the expression of NOR1 in mRNA and protein levels. Further research should focus on the role of NOR1 expression in endothelial injury and atherosclerosis inflammation.
