摘 要:胰岛移植被认为是治疗1型糖尿病的理想方式。然而,胰岛供体来源缺乏限制了该技术在临床的广泛应用。更多的研究者将目光转向异种胰岛移植。猪作为首要替代胰岛来源,面临的异种排斥反应比同种移植更为剧烈,传统的免疫抑制策略无法满足长期脱离胰岛素的要求。近几年基因修饰技术不断进步,推动了改良基因猪胰岛在异种胰岛移植方面的发展,并取得显著成果。本文就通过基因修饰改良供体猪胰岛使其能在受体内长期存活保持功能的研究进展进行综述。Islet transplantation is considered as a prospective treatment for patients with type 1 diabetes mellitus (T1 DM ). However, the shortage of available islet grafts greatly restricts the further application of clinical islet transplantation. Therefore, more researchers have pail attention to the investigation on xenogeneic islet transplantation. For xenotransplantation the pig is promising candidate, immune rejection is more violent than allotransplantation, and the common immunosuppressive strategy cannot meet the needs of xenotransplantation. In recent years, great progress in gene modification technology has promoted the development of modified porcine islet in xenotransplantation and achieved remarkable results. In this paper, we reviewed the progress of research on improving the long - term survival and maintaining function of donor porcine islets in xenotransplantation through gene modification.
