摘 要:为探明绵羊虱蝇雌、雄成虫中肠及蛹的菌群结构特点,提供防控绵羊虱蝇及其传播疾病的科学依据,本研究在无菌条件下采集雌、雄绵羊虱蝇成虫中肠内容物及蛹内容物,用试剂盒提取细菌总DNA,运用通用引物PCR扩增细菌16S r DNA V3区,DGGE电泳,选取优势条带,回收、测序,测序结果与Gen Bank数据库中已知序列进行比对。结果显示,雌、雄绵羊虱蝇成虫中肠及蛹的优势菌均为巴尔通氏体、杀雄菌、沃尔巴克氏体。研究结果表明,雌、雄绵羊虱蝇成虫中肠及蛹的菌群结构相似,且巴尔通氏体、杀雄菌、沃尔巴克氏体可垂直传播。The characteristics of microflora in the midgut and pupa of Melophagus ovinus were analyzed by using denaturing gradient gel electrophoresis(DGGE).The samples of the midgut and pupa contents were collected under aseptic conditions,total DNA of bacteria was extracted according to the manufacturer's protocol,and the universal primers were used to amplify the bacteria of V3 areas of 16 S rDNA.The predominance bands were cut for recovery and sequencing after DGGE,and sequencing results were compared with the known sequences in Gen Bank.In result,the dominant bacteria in the midgut and pupa of M.ovinus were Bartonella,Arsenophonus and Wolbachia. The results indicated that microflora in the midgut of female and male adults of M.ovinus were similar to the pupa,Bartonella,Arsenophonus and Wolbachia,and had capability to vertical transmission.
