摘  要:2016年4月,四川邛崃某鸭场送检30只樱桃谷肉鸭,临床表现鸭喙变短、舌头伸出。从送检鸭脏器中分离鉴定出1株鸭源细小病毒,命名为QH-L01株。为进一步了解该病毒毒力及分子生物学特性,对其进行ELD50测定、VP1基因测序,并与其他细小病毒毒株进行比较。ELD50测定结果为10-6.54ELD50/0.2 m L;VP1核苷酸和氨基酸序列分析结果表明,该病毒结构基因全长2 203 bp,编码734个氨基酸,与鹅细小病毒核苷酸同源性为92.8%~99.8%,与番鸭细小病毒同源性为77.4%~87.3%;VP1编码的氨基酸同源性和鹅细小病毒及番鸭细小病毒分别为95.2%~99.5%和85.4%~91.1%。VP1基因进化分析显示,该病毒和鹅细小病毒SYG26-35分离株处于同一进化分支上。动物回归试验结果显示,试验组出现明显的鸭源细小病毒BADS症状,对照组无明显变化,表明该病毒分离鉴定成功。In April,2016,samples were collected from duck with beak atrophy and dwarfism syndrome on a duck farm in Qionglai,Sichuan Province.A Cherry Valley duck-origin parvovirus was isolated and named QH-L01 strain.In order to further understand the molecular characteristics and virulence of this virus,ELD50 was determined,the VP1 gene was sequenced and compared with that of other parvovirus strains.The result of ELD50 was 10-6.54ELD50/0.2 mL.Nucleotide and amino acid analysis showed that the structural VP1 gene was 2203 bp in size,encoding for a protein of 734 amino acids,and the QH-L01 isolate VP1 gene shared 92.8% to 99.8% identity and 77.4%—87.3% with that of GPV and MDPV,respectively.The mino acids shared 95.2%—99.5% and 85.4%—91.1% identity with that of GPV and MDPV,respectively.The results of animal experiment showed that there were obvious symptoms of duck-origin parvovirus BADS in the experimental groups,and there were no changes in the control groups,indicating the successful virus isolation and identification.
