摘 要:目的分离和鉴定具有抗菌活性的红树林根际土壤放线菌,以期发现具有药用价值的放线菌资源。方法采用4种分离培养基以稀释涂布法分离放线菌,对分离自红树林根际土壤放线菌进行形态特征观察、生理、生化和和抗菌活性测定;采用PCR技术测定16SrRNA基因序列,进行比对分析和系统发育分析。结果从红树林根际土壤样品中分离出6株典型放线菌,其中BLHK5、BLHK125、BLHK198、BLHK245、BLHK271具有广谱抗菌活性,抑菌圈直径为926mm;根据其形态特征、生理生化和基于16SrRNA系统发育分析,初步鉴定BLHK125、BLHK271为小单孢菌属(同源性分别为99.83%和98.53%),BLHK217为糖单孢菌属(同源性为98.88%),BLHK5、BLHK198、BLHK245为链霉菌属(同源性为99.10%100.00%)。结论广西北部湾北仑河口红树林根际土壤放线菌资源丰富,从红树林根际土壤样品中分离的5株放线菌的发酵液浓缩物具有较好的抗菌活性,具有研究价值。Objective To isolate and identify Actinomycetes strains with antimicrobial activity from mangrove rhizosphere soil in order to identify actinomycetes with medicinal value. Methods Actinomycetes strains were isolated from mangrove rhizosphere soil using dilution plating with four separate cultures.The morphology of the strains was observed,the strains were biochemically identified,and their antimicrobial activity was assayed.The sequence of the 16 SrRNA gene was determined using PCR,sequences were compared,and a phylogenetic analysis was performed. Results Six typical strains of Actinomycetes were isolated from mangrove soil samples.Five of the six-BLHK5,BLHK125,BLHK198,BLHK245,and BLHK271-had broad-spectrum antibacterial activity,with an inhibition zone diameter ranging from 9to 26 mm.The morphology,physiological properties,biochemical properties of the strains and sequences of the16 SrRNA gene suggested that the BLHK125 and BLHK271strains of Actinomycetes were Micromonospora(similarity of99.83% and 98.53%,respectively),the BLHK217 strain s was Saccharomonospora(similarity of 99.88%)and the BLHK5,BLHK198,and BLHK245 strains were Streptomyces(similarity ranged from 99.10%to 100.00%). Conclusion Five Actinomycetes strains were isolated from mangrove rhizosphere soil in the Beilun estuary,Guangxi,and their fermentation products displayed significant antibacterial activity.Mangrove rhizosphere soil is rich in culturable medicinal actinobacteria,warranting further research.
