摘  要:杜仲降血压的主要成分是松脂醇二葡萄糖苷(PDG).采用表面灭菌法从杜仲枝条、树皮中分离得到3株可以产生松脂醇二葡萄糖苷(PDG)的内生真菌,其中菌株YH001通过形态观察和ZTS序列分析,初步鉴定为茎点霉属菌株(Phomopsis),摇瓶培养检测发酵液中PDG产量后发现菌株YH001的PDG产量可以达37.98mg/L.通过Plackett-Burman试验设计,确定YH001的培养基中的显著影响因素为绿豆粉、葡萄糖和硝酸钠的含量,对液体培养条件进行优化,最终菌株YH001PDG产量可达75.56mg/L.Pinoresinoldiglucoside (PDG) is the main antihypertensive component in Eucommia ul- rnoides Oliv.By surface sterilization,we isolated 3 strains of endophytes fungi which could produce PDG from the branch and bark of Eucommia ulmoides Oliv. It was confirmed that the 3 strains of endophytes fungi were Phomopsis through morphology comparison.The yield of PDG could reach 37.98 mg/L when these strains were cultivated in liquid medium in flask.Through the Plackett-Burman test design, we con- firmed that the respect content of mung bean powder,glucose and NaNO3 inmedium had great influence on the production of PDG.The final PDG concentration could reach 75.56 mg/L through optimizing the liquid culture conditions.
