摘 要:[目的]探讨蓝猪耳花粉管的最佳培养条件,为建立植物离体受精体系奠定基础。[方法]采用离体培养技术,研究了蓝猪耳花粉离体萌发和花粉管体外生长的主要影响因素。[结果]蓝猪耳花粉萌发及花粉管生长的最佳培养基为:5.00%(V/V)蔗糖、0.01%(V/V)氯化钙、0.01%(V/V)硼酸和0.01%(V/V)磷酸二氢钾,其培养基的渗透压主要靠蔗糖调节。[结论]硼是蓝猪耳花粉萌发中必不可少的微量元素;钙对花粉管生长起着非常重要的调节作用,并推测钙离子主要是通过电压依赖型钙通道进入花粉管内部。[ Objective ] To explore the optimum culture conditions for the pollen tube of Torenia fournieri. [ Method ] Using the techniques of in vitro culture, the main influencing factors of pollen germination and pollen tube' s growth in vitro of T. fournieri were studied. [ Result] The suitable culture medium for pollen germination and pollen tube' s growth in vitro of T. fournieri was as follows:5% (V/V) sucrose, 0.01% (V/V) CaC12, 0.01% (V/V) boric acid and 0.01% (V/V) KH2po4, The osmotic pressure of the medium was mainly regulated by sucrose. [ Conclusion] Boron is an essential microelement in pollen germination of T. fournieri. Calcium plays an important role in pollen tube' s growth. It is deduced that voltage-dependent Ca^2+ channels may be a main calcium influx pathway through which Ca^2+ entered into pollen tube.
