摘  要:目的探讨在冰冻红细胞去甘油化时添加适量的蔗糖溶液,提高红细胞回收率的可行性。方法选择6天内制备的400 ml去白悬浮红细胞60袋,制备成冰冻红细胞,置于-80℃^-65℃的冰箱内保存,在保存4个月后取出解冻去甘油化,并随机分成实验组和对照组,每组各30袋。实验组在去甘油化过程中的第1次洗涤液改用10%的蔗糖溶液,第2次洗涤液改用10%的蔗糖溶液,对照组仍用0.9%的氯化钠溶液进行洗涤,然后分别比较两组的红细胞回收率,甘油残留量和去甘油化过程中的各个洗涤阶段的游离血红蛋白含量。应用SPSS13.0软件,所获数据采用方差分析、t检验。结果两组红细胞回收率比较,P<0.0005;,甘油残留量比较,P>0.05。两组去甘油化第1次洗涤比较,P<0.0005;第2次洗涤、第3次洗涤、第4次洗涤比较,P>0.05。结论冰冻红细胞去甘油化过程中第一阶段的洗涤过程非常重要,对红细胞进行保护,可以显著提高红细胞回收率。Objective To explore the frozen red blood cells when to glycerol to add just the right amount of sucrose solution, increasing the recovery ratio of red blood cells. Methods Choose 400 ml of the preparation of 6 days to 60 bag white suspended red blood cells, preparation of frozen red blood cells, under -80℃--65℃ refrigerator.save, save 4 months later to take out the thaw to glycerol, and randomly divided into experimental group and control group, each group 30 bags. Experimental group in the first washing liquid in the process of use to glycerin 10% sucrose solution, second washing liq-uid to switch to 10% sucrose solution, the control group still washing with 0.9% sodium chloride solution, and then respectively to compare two groups of red blood cells, recovery of glycerin residues and to glycerol in the process of free hemo- globin content in all stages of washing.Using SPSS13.0 software, the data obtained using analysis of variance and t test. Results Two groups of red blood cells recovery, delta P〈0.0005; Residue, glycerin, P〉0.05. Two groups to glycerol first washing comparison, P〈0.0005. Second washing, 3 times of washing, 4 times of washing, P〉 0.05. Conclusion Frozen red blood cells to glycerin is changed in the process of the first stage of the washing process is very important, to protect the red blood cells can significantly improve erythrocyte recovery.
