摘  要:目的:研究苦瓜总皂苷对IgA肾病大鼠转化生长因子-β(Transforming growth factor-β,TGF-β)表达的影响。方法:随机选取48只雄性SD大鼠,所有大鼠均于适应性喂养1周后随机分为空白对照组、IgA肾病组以及苦瓜总皂苷治疗组,n=16,经典造模法造模成功后,治疗组给予苦瓜总皂苷20 mg/(kg·d)治疗,IgA肾病组和空白组给予等量生理盐水灌胃1 m L/(kg·d),分别在治疗4周和8后,每组处死一半大鼠,观察血清尿素氮、血肌酐、24 h蛋白尿和血清中白介素-2(IL-2)的白介素-6(IL-6)的含量,此外HE染色分析其病理学,Western Blot和RT-PCR检测TGF-β_1和RGC-32表达。结果:IgA肾病组和治疗组所有大鼠均造模成功,主要表现为有明显肉眼或镜下血尿;此外与空白组相比,IgA肾病组和治疗组血清中BUN、24 h尿蛋白、SCr、IL-2和IL-6明显升高(P<0.05),苦瓜总皂苷治疗4周后,上述指标均明显降低(P<0.05),治疗8周后进一步降低;病理学检查显示空白组纹理结构清晰,而IgA肾病组有明显增生反应,苦瓜总皂苷对其有一定的改善。Western Blot和RT-PCR结果显示与空白组相比,IgA肾病组肾脏组织中TGF-β_1表达明显降低(P<0.05),治疗8周后进一步降低,但其对RGC-32基本没有改善作用。结论:苦瓜总皂苷可通过降低肾脏组织中TGF-β_1的表达,进而有助于炎性因子和肾功能的改善,防止其疾病的进一步进展。Objective:To study and analysis the affection of the total momordicoside to TGF-β_1 in IgA nephropathy in rats. Methods:48 male SD rats were randomly divided into the normal group,IgA group and total momordicoside treatment group,n=16,classic building method was adopted to built IgA nephropathy rats model,after the success of the building,total momordicoside treatment group was given total momordicoside 20 mg/(kg·d)lavage treatment,respectively,the IgA group and normal group was given the corresponding amount of saline 1 m L/(kg·d),after treatment 4 and 8 weeks,to observe the rat serum creatinine and urea nitrogen,24-hour proteinuria serum,IL-2 and IL-6 levels. HE staining detected the pathology,and Western Blot,RT-PCR to detected the TGF-β_1 and RGC-32 expression. Results:After the successful built model,the IgA group and the treatment group had obvious naked. Compared with the normal group,the IgA group in 24 h urinary protein,serum Cr,BUN and IL-2 and IL-6 were significantly elevated,using total momordicoside to treated 4 weeks except the Cr,other indicators were significantly lower,after 8 weeks treatment the above five indexes were significantly reduced;the results of HE staining showed the normal group was clear,and the IgA group had obvious proliferation response,after using total momordicoside treated,it had certain improvement. Western Blot and RT-PCR results showed that,compared with the blank group,IgA group TGF-β_1 was significantly elevated,and it were significantly lower after treatment using total momordicoside(P〈0.05). Conclusion:Total momordicoside can by lowering the serum IL-2 and IL-6 levels in IgA nephropathy of rats,thereby inhibiting TGF-β_1 expression,eventually improve the symptoms of rats.
