摘  要:为了探讨α干扰素(IFN-α)对体外培养猪黄体细胞增殖及VEGF基因表达的影响,试验分离猪卵巢中的黄体组织进行消化处理,制成细胞悬液,体外培养猪黄体细胞,采用MTT比色法测定猪黄体细胞的增殖,ELISA法测定培养液中孕酮的浓度;Real time PCR和Western-blot检测血管内皮生长因子(VEGF)基因表达。结果表明:IFN-α对猪黄体细胞增殖的抑制作用随着浓度升高而增强。在IFN-α浓度低于100 IU/m L时,对猪培养黄体细胞分泌孕酮呈现抑制作用;当IFN-α浓度高于100 IU/m L时,对猪培养黄体细胞分泌孕酮呈现促进作用。IFN-α下调猪黄体细胞中VEGF mRNA和VEGF蛋白表达。说明IFN-α抑制黄体细胞增殖并具有浓度依赖性,对猪黄体细胞中孕酮的分泌没有影响,下调猪黄体细胞中VEGF基因的表达。To investigate the effects of IFN - α on the proliferation of sow luteal cells and the expression of VEGF in vitro. The corpus luteum was isolated from the ovary of sow, and the tissue was digested to prepare cell suspension. The luteal ceils were cultured in vitro. The proliferation of luteal ceils was deter mined by MTT colorimetry. The concentration of progesterone in culture medium was deter mined by ELISA. Real time PCR and Western - blot were used to detect VEGF gene expression. The results showed that the inhibithion of IFN - α on the proliferation of sow luteal cells increased with concentration. When the sow cultured luteal cells secreting progesterone displayed inhibition, IFN -α concentration was below 100 IU/mL. When IFN - α concentration is higher than 100 IU/mL,IFN - α promotes the secretion of progesterone from sow cultured luteal cells ; IFN - α downregulated the mRNA and protein expression of VEGF in sow luteal cells. It was concluded that IFN - a inhibited the proliferation of luteal cells in a concentration dependent manner. There was no effect on progesterone secretion of sow luteal cells. The expression of VEGF was dcreased by IFN -α in sow luteal cells.
