马骨关节炎模型关节液中相关生物指标变化的研究

摘　　要：为了研究马骨关节炎发病机理,试验将13匹蒙古马分成两组,试验组8匹(用两性霉素B诱导并建立马骨关节炎模型),对照组5匹,每周采集1次关节液,直至第9周,用ELISA方法检测关节液中前列腺素E2(PGE_2)、肿瘤坏死因子-α(TNF-α)、基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)的含量。结果表明:试验组关节液中MMP-2、MMP-9、TNF-α和PGE_2的含量有增加的趋势,其中MMP-9、PGE_2在发病初期就显著上升。说明在骨关节炎过程中,关节液中MMP-2、MMP-9、PGE_2、TNF-α都呈上升趋势,这些促炎因子的增加会促进蛋白酶的分泌和软骨的降解。In order to study the pathogenesis of osteoarthritis, thirteen Mongolian horses were divided into two groups. The experimental group consisted of 8 horses （ induction and establishment of equine osteoarthritis model with amphotericin B）. The control group consisted of 5 horses. The contents of prostaglandin E2 （ PGE2 ）, tumor necrosis factor - α （TNF - α）, matrix metalloproteinase - 2 （ MMP - 2 ）, matrix metalloproteinase - 9 （ MMP - 9） in the synovial fluid were measured by ELISA weekly. The experimental period was nine weeks. The results showed that the levels of MMP - 2, MMP - 9, TNF - α and PGE2 in the synovial fluid of the experimental group were increased, MMP - 9 and PGE2 were increased significantly in the early stage. It was concluded that MMP - 2, MMP - 9, PGE2 and TNF - α in synovial fluid were increased in the process of osteoarthritis, and the increased proinflammatory cytokines could promote the secretion of pretease and cartilage degradation.
