摘 要:目的:探讨利拉鲁肽对2型糖尿病(type 2diabetes mellitus,T2DM)大鼠血糖和Lee’s指数的影响及重要靶器官的保护作用。方法:成年SD大鼠高脂喂养12周后腹腔一次性注射2%链脲佐菌素(streptozotocin,STZ)30mg/kg,建立T2DM模型。模型建立后,利拉鲁肽大、小剂量组分别给予利拉鲁肽0.2mg/kg、0.1mg/kg,皮下注射共4周。动态观察大鼠体重、体长、血糖等变化,测定Lee’s指数。第16周末取大鼠肺、肾、胰腺、视网膜组织行HE染色观测形态学变化。结果:成模4周末,利拉鲁肽大、小剂量组大鼠Lee’s指数分别为311.8±6.4和317.6±4.3,明显低于DM组的324.2±7.2,差异有统计学意义(P<0.05)。DM组大鼠血糖水平持续升高,与DM组比较,利拉鲁肽组血糖水平显著下降(P<0.05),且大剂量组效果更明显。HE染色显示利拉鲁肽干预后糖尿病大鼠的肺、肾和胰腺组织病理损害较DM组明显减轻。各组大鼠视网膜未见明显病理改变。结论:利拉鲁肽可有效减低糖尿病大鼠血糖和Lee’s指数,并能明显改善糖尿病所致的肺、肾、胰腺等重要脏器的病理损伤。Objcctive:To investigate the effect of liraglutide on plasmal glucose level and I-ee,s index and its protective role on the lesion of important target organs in rat model with type 2 diat〉etes mellitu.s(T2DM). Methods:The T2DM rat model was evStablished by feeding high-fat diet for 12 weeks and followed by intraperitoneal injection of streptozotocin (STZ) in a single dose of 30 mg/kg. Then the rats in liraglutide group were administered by intramuscular injection twice daily in a dose of 0. 1 mg/kg or 0. 2 mg/kg for 4 weeks. The plasma glucose level,the body weight and length were 〇 bsen ,ed dynamically, and the I-ee’ s index was calculated. HE staining of lung, kidney, retina and pancreatic tissues were performed to observe the histopathologic change. Resul ts: At the end of the fourth week after modeling,the I^ee,s index in low-dose and high-dose liraglutide groups were 311. 8±6. 4 and 317. 6±4. 3,respectively,which were obviously lower than 324. 2i7. 2 in DM group* *and the difference was statistically significant (P〈0. 05). The plasma glucose level was significantly increased in DM group;compared with DM group,liraglutide administration obviously reduced the plasma glucose level (P 〈C〇 . 05),which was more prominent in high-dose liraglutide group. HE staining showed that the histopathologic lesion of lung, kidney and pancreatic tissues in diabetic rats was obviously alleviated after liraglutide intervention. The retinal histopathologic lesion was not prominent in each group. Conclusion: Liraglutide treatment can effectively decrease Lee's index and plasma glucx)se level in type 2diabetic rats,and significantly improve histopathologic lesion of lung, kidney and pancreatic tissues induced by diabetes.
