摘 要:目的从人眼袋脂肪组织中分离获取脂肪来源间充质干细胞(MSCs),观察比较其生物学特性。方法以眼睑美容术中废弃的眼袋脂肪组织为材料,皮下脂肪组织为对照,胶原酶消化法分离脂肪组织来源的MSCs,常规方法加以培养。倒置显微镜观察MSCs的形态特征,采用MTT法绘制细胞生长曲线,并用流式细胞仪分析细胞表面分子标志物,同时进行成骨、成脂诱导和细胞衰老特性的观察。结果从人眼袋脂肪中分离获取了与皮下脂肪来源相同形态和生长特征的MSCs,人眼袋脂肪来源的MSCs表达CD105、CD166,不表达CD34、CD54,油红O染色和茜素红染色证实其可进行成骨、成脂诱导,第5代开始出现细胞衰老的表现。结论可从眼睑美容术中废弃的眼袋脂肪中有效分离获取脂肪来源MSCs,具有较好的生物学特性。Objective The purpose of this study was to isolate human adipose derived stem cells from orbital fat and to observe their biological characteristics. Methods Mesenchymal stem cells was isolated from human orbital fat by a collagenase digestion method compared with subcutaneous fat. Cellular morphology was observed with inversion microscopic. Cell growth kinetics was obtained by MTT method. The phenotype of isolated human orbital fat derived stem cells was analyzed by flow cytometry. The biological characteristics of orbital fat-derived stem cells were examined by osteogenic and adipogenic differentiation as well as β-Galactosidase staining. Results Ours results showed that the mesenchymal stem cells was isolated from human orbital fat with a similar cellular morphology and growth kinetics that of mesenchymal stem cells from subcutaneous fat. The orbital fat-derived stem cells expressed CD105、CD166,not CD34、CD54. It is showed orbital fat-derived stem cells had the peculiarity of osteogenic and adipogenic differentiation by oil red O staining and alizarin red staining. Cell senescence of orbital fat-derived stem cells was observed in 5th subculture. Conclusion These findings suggested the mesenchymal stem cells could acquired from discarded orbital fat of cosmetology with a well biological characteristic.
