摘  要:【目的】测定青海产的高海拔植物蕨麻根中α-生育酚的含量。【方法】样品经KOH皂化后,采用正己烷和乙酸乙酯混合溶剂提取,以甲醇-乙腈(30:70,v/v)为流动相,经HPLC-荧光法测定α-生育酚含量。【结果】α-生育酚在1~16μg/ml质量浓度范围内呈良好的线性关系,日内相对标准偏差为1.18%~1.45%,日间相对标准偏差为1.40%~1.87%,日内重现性RSD为2.71%,日间重现性RSD为3.84%,方法回收率为90.08%~92.25%。青海产蕨麻中α-生育酚含量为81.25~84.47μg/g。【结论】建立的HPLC-荧光分析方法测定蕨麻根中α-生育酚的含量,方法灵敏、准确,提取效率高。【Objective】To determine the concentration of α-tocopherol in Potentilla anserina L with origin of Qinghai.【Methods】After saponification in KOH solution, the sample was extracted using solvent mixture of n-hexane and ethyl acetate. With methanol:acetonitrile(30:70) as the mobile phase, the concentratins of α-Tocopherolin the extracts was determined by HPLCcombinated with a fluorescence Detector.【Results】The concentration ofα-tocopherol showed good linearity in the range of 1-16 μg/ml. The RSDs of intra-day and inter-day precision were1.18%-1.45% and 1.40-1.87%. The RSDs of intra-day and inter-day repeatabilitywere 2.71%and 3.84%.The recovery rates indifferent concentrations were 90.08%-92.25%. The amount of α-tocopherol in the Potentilla anserina L was 81.25-84.47 μg/g.【Conclusion】This established analysis method is sensitive, accurate and high efficiency of extraction, suitable for determination ofthe α-Tocopherol concentrationsinthe root of Potentilla anserine L.
