摘 要:【目的】研究不同凝胶基质对凝胶剂中左旋肉碱体外经皮渗透的影响,并评价凝胶剂作为左旋肉碱透皮给药载体的可行性。【方法】制备不同基质的左旋肉碱凝胶,采用改良的Franz扩散池,用离体大鼠皮肤进行体外经皮渗透实验,以HPLC法检测接受液中左旋肉碱含量并计算累积渗透量及稳态透皮速率。【结果】以2%羧甲基纤维素钠为基质的左旋肉碱累积透过量最大(7.635 mg/cm^2),透皮速率常数(J)为0.2308 mg·(cm^2·h)^(-1),均优于甲基纤维素和卡波姆。【结论】选用2%羧甲基纤维素钠作为左旋肉碱凝胶剂的基质较好。【Objective】To study the effects of different gel matrices on the in vitro transdermal permeability of L-carnitine, and evaluate the feasibility of transdermal delivery carriers containing L-carnitine.【Methods】The different gels containing L-carnitine were formulated. The transdermal permeation test was carried out by using improved Franz diffusion cell through in vitro rat skin. The concentration of L-carnitine in receptor medium was measured by HPLC. Then the cumulative penetration amount and steady transdermal penetration rate were calculated.【Results】The cumulative penetration amount of the gel was the most(7.635 mg/cm^2)when the matrix was 2% carboxy methyl cellulose sodium(CMC-Na). The penetration rate constant(J) was 0.2308 mg·(cm^2·h)^-1. The indexes of CMC-Na were better than those of methyl cellulose(MC) and carbomer.【Conclusion】The optimum matrix for L-carnitine transdermal system is 2% CMC-Na.
