民族药地板藤中芦丁TLC鉴别与HPLC含量测定研究

摘　　要：目的:建立民族药地板藤药材中芦丁的TLC鉴别与HPLC含量测定方法,为完善地板藤质量标准提供参考。方法:采用TLC法对地板藤中的芦丁进行定性鉴别;采用HPLC法对地板藤中芦丁进行含量测定,以Diamonsil C_(18)(250 mm×4.6 mm,5μm)为色谱柱,流动相为0.2%磷酸水-乙腈(70∶30)等度洗脱,流速为0.8 m L·min^(-1),检测波长为360 nm,柱温为35℃。结果:药材中芦丁成分在荧光下薄层色谱斑点清晰,专属性强;芦丁在1.07~10.7μg范围内线性关系良好(r=0.9993),平均回收率为100.1%,RSD%为1.12%(n=5)。结论:所选指标为地板藤药材中的特征成分,所建方法快速、简便、准确、重复性好,可作为地板藤的质量控制方法。Objective： To establish an effective TLC and HPLC method for qualitative and qualitative anal- ysis of rutin in Ficus tikoua Bur. which can be used to be one of the quality control of Ficus tikoua Bur.. Methods： Rutin was identified by TLC in Ficus tikoua Bur.; Rutin was determined bu RP-HPLC. Chromatogra- phy conditions were Diamonsil C18 （2） （250 minx4.6 ram, 5 μm） column with 0.2% phosphoric-acetonitrile （70：30） as mobile phase; the flow rate was 0.8 mL＇min-l; UV detection wavelength was set at 360 nm, and the column temperature was 35 ℃. Results： The TLC sports developed were fairly clear with no interference; linear correlation of the peak area and the concentration of Rutin over the concentration range 1.07-10.7 μg was obtained （r= 0.9993）. The average recovery was 100.1% and RSD was 1.12% （n=5）. Conclusion： Rutin was characteristic compound in Ficus tikoua Bur.. The method is simple, rapid and accurate, which can be used for the quantity control method of Fieus tikoua Bur..
