摘  要:目的:探讨海藻糖对糖氧剥夺所致SH-SY5Y细胞损伤的保护作用及可能机制。方法:体外培养人SH-SY5Y细胞,建立糖氧剥夺损伤模型。通过乳酸脱氢酶(LDH)释放、锥虫蓝染色来检测细胞存活情况;采用DCFH-DA染色检测细胞内活性氧含量。结果:与对照组相比,海藻糖不仅能够明显抑制缺氧缺糖(OGD)条件下的神经细胞死亡(P<0.05),还能抑制过氧化氢所致SH-SY5Y细胞死亡。结论:海藻糖对糖氧剥夺所致SH-SY5Y细胞损伤的保护作用,其机制可能与其抑制氧化应激反应有关。Objective To investigate the protective effect of trehalose on SH-SY5Y cell injury induced by oxygen glucose deprivation(OGD) and its possible mechanism. Methods Human SH-SY5Y cells were cultured in vitro to establish a model of glucose deprivation injury. Cell viability was detected by lactate dehydrogenase (LDH) release and trypan blue staining, and the intracellular reactive oxygen speeies(ROS) was measured by DCFH-DA staining test. Results Compared with the control group, trehalose can not only inhibit the neuronal death under OGD condition (P 〈0.01), but also inhibit the death of SH-SY5Y cells induced by hydrogen peroxide. Conclusion The protection by trehalose against SH-SY5Y cell damage caused by OGD is possibly related to its inhibition of oxidative stress.
