摘  要:为了探究庆大霉素致原代肾小管上皮细胞的凋亡作用,以0、1、2、4、8、16 mmol/L浓度的庆大霉素分别作用原代大鼠肾小管上皮细胞24 h,采用cck-8法检测细胞相对活力,hochest 33258染色法观察细胞核形态以及利用流式细胞术检测细胞凋亡率。结果表明,与对照组相比,随着庆大霉素浓度的增加,细胞活性呈显著或极显著下降(P<0.05或P<0.01),细胞凋亡率呈显著或极显著上升(P<0.05或P<0.01),hochest33258染色观察到细胞核出现固缩或碎裂(P<0.01)。结果提示,庆大霉素能引起细胞发生凋亡和损伤,并且这可能与其肾毒性有密切的联系。To investigate the apoptosis effects of gentamicin on primary culture of renal epithelial cells,primary culture of renal epithelial cells were treated with gentamicin( 0 ~ 16 mmol/L) for 24 h. CCK-8 assay was used to determine the relative viability of cells. Flow cytometry( FCM) and hochest 33258 staining were used to detect apoptotic rate. The results showed that compared with the control group,the relative viability of cells was decreased and the apoptotic rate increased( P〈0. 05 or P〈0. 01),pyknosis or chromatin condensed after hochest33258 staining as the( P〈0. 01). The results suggested that gentamicin-induced cell apoptosis and maybe connected with gentamicin-induced renal toxicity.
