摘 要:探究不同碳源、氮源、微量元素、pH、温度等对玉米黑粉菌菌丝生长的影响,选出适合玉米黑粉菌生长的条件。通过单因素试验对菌丝形态、生长势、生长速度进行分析比较,选出适合玉米黑粉菌生长的最佳碳源、氮源、微量元素、pH和温度。通过正交试验优化碳源、氮源、微量元素的比例。结果表明,最佳生长条件为葡萄糖1%、酵母粉1%、MgSO_4 0.1%、KH_2PO_4 0.15%,pH6~7,温度20℃~30℃。培养周期为4 d^7 d。通过对玉米黑粉菌的生物学特性研究,掌握其菌丝体生长、繁殖所适宜的营养条件及环境条件,为玉米黑粉菌的开发利用提供参考。Effects of different carbon sources, nitrogen sources, trace elements, pH value and temperature on mycelial growth of Ustilago maydis were studied, and the conditions suitable for the growth of U. maydis were selected. The optimal carbon source, nitrogen source, trace element, pH and temperature were selected for the growth of strain by single factor investigation. The pro- portion of the carbon source, nitrogen source, trace elements were optimized by orthogonal experiment. The optimum growth conditions were glucose of 1%, yeast powder of 1%, MgSO4 of 0.1%, KH2PO4 of 0.15%, pH of 6-7, temperature of 20℃-30℃. The incubation period is 4 d-7 d. The biological characteristics of U. maydis were studied to understand the suitable nutrient conditions and environmental conditions of mycelium growth and reproduction, and to provide reference for the development and utilization of U. maydis.
