摘  要:目的:探讨生发片组方水提物对小鼠B16黑素瘤细胞酪氨酸酶活性和黑素合成的影响及作用机制。方法:采用MTT法筛选合适的药物浓度,L-多巴氧化法检测酪氨酸酶活性,氢氧化钠裂解法检测黑素生成,Western blotting检测MITF、TYR及p-CREB蛋白的表达水平。结果:与对照组相比,在0.25~1g·L-1浓度范围内,生发片组方水提物可明显促进酪氨酸酶活性和黑素合成,上调MITF、TYR及p-CREB蛋白的表达。结论:生发片水提物可能通过PKA信号通路促进黑素的合成。Objective:To research the effect and mechanism of sheng fa tablet aqueous extract on tyrosinase activity and melanogen- esis in B16 melanoma cells. Methods: The appropriate concentration screened with MTT; The tyrosinase activity was detected by L- DOPA-Oxidation;the amount of melanin was detected by NaOH dissolvedmethod;the expression of MITF, TYR and p-CREB was detected by Western blotting. Results:Range 0.25g·L-1 to 1g·L-1 ,the tyrosinase activity,the amount of melanin and the expression of MITF,TYR and p-CREB in sheng fa tablet group were higher than those in control group. Conclusion:Sheng fa tablet can promot the melanogenesis in B16 melanoma cells through the PKA pathway.
