摘 要:以成熟红豆果为外植体,在新成熟红豆果外壳生长点处进行机械剪口破壳处理后,接种于经高压灭菌的种子胚芽诱导培养基WPM+6-BA4.44μmol/L+NAA0.54μmol/L+Su 3%+AC 0.15%+Ag 0.65%中,待种胚芽长至5~6 cm时,切取芽的顶部、中段分别作为扩大繁殖的繁殖体,余下的子叶部分采用机械处理方式诱导潜伏芽的形成.适宜下胚轴芽诱导培养基为WPM+6-BA 4.44μmol/L+NAA 0.54μmo儿+TDZ 0.05μmol/L+Su 3%+AC 0.1 5%+Ag 0.65%,芽诱导率约达80%;适宜不定芽诱导培养基为WPM+6-BA 4.44μmol/L+TDZ 0.05μmol/L+NAA 1.61μmol/L+KT 2.23μmol/L+Su 3%+AC 0.15%+Ag 0.65%;适宜壮苗培养基为WPM+6-BA2.22μnol/L+NAA 1.6lμmol/L+KT 2.23μmol/L+Su 3%+AC 0.15%+Ag 0.65%;适宜生根培养基为1/2WPM+IBA 4.92μmol/L+NAA 2.67μmol/L+Su 3%+AC 0.15%+Ag 0.65%.成功诱导下胚轴潜伏芽萌发并形成完整植株,为木本植物红豆树高效繁殖提供了一个新途径和有效方法.Using mature Ormosia hosiei fruits as explants, the fruit shell was broken by mechanical cutting at the shell growing point. These explant materials were placed onto autoclaved induction medium for seed em- bryo: WPM+ 6-BA 4.44 μmol/L + NAA 0.54 μmol/L+ Su 3% +AC 0.15% + Ag 0.65%. When embryos were grown to a length of 5-6 cm, the top and middle segments of the stems were cut. The epicotyl part was used as propagules to increase reproduction and the remaining part of cotyledons was used to induce germination of plant organ latent buds of hypocotyls with mechanical treatment. The optimal medium for hypocotyl bud induc- tion was WPM+ 6-BA 4.44μmol/L + NAA 0.54 μmol/L + TDZ 0.05 μmol/L + Su 3% +AC 0.15% + Ag 0.65% and the obtained induction rate of bud was 80% under this condition. The most suitable medium for from hypo-eotyls was WPM + 6-BA 4.44μmol/L+ TDZ 0.05μmol/L+ NAA 1.61 μmol/L+ KT 2.23μmol/L+ Su 3% + AC 0.15% + Ag 0.65%. The optimal medium for seedling strengthening was WPM+ 6-BA 2.22 μmol/L+ NAA 1.61 μmol/L+ KT 2.23 μmol/L+ Su 3% + AC 0.15%+ Ag 0.65%. The optimal medium for rooting was 1/2WPM + IBA 4.92 μmol/L + NAA 2.67 μmol/L + Su 3% + AC 0.2% + Ag 0.65%. The cotyledon hypocotyl latent buds were successfully induced and propagated. This method provide a new and effective way of industrial propa- gation for Ormosia hosiei.
