摘  要:目的观察人参皂苷对波动性高糖所致内皮细胞损伤的保护作用并探讨其作用机制。方法体外培养人脐静脉内皮细胞(HUVECs),分为对照组、波动性高糖模型组及人参皂苷低中高浓度组(12.5μg/mL、25μg/mL、50μg/mL)。Western blot检测NF-E2相关因子2(Nrf2)的核转位及下游抗氧化蛋白血红素加氧酶-1(HO-1)、γ-谷氨酰半胱氨酸合成酶(γ-GCS)和醌氧化还原酶(NQO1)的表达;RT-PCR检测下游抗氧化蛋白HO-1、γ-GCS和NQO1的m RNA表达;Western blot检测瞬时转染Nrf2 si RNA质粒后Nrf2总蛋白和核蛋白及下游HO-1、γ-GCS和NQO1蛋白的表达。结果人参皂苷治疗后,HUVECs中的Nrf2核蛋白及下游抗氧化蛋白HO-1、γ-GCS和NQO1表达均明显增强(P<0.05)。瞬时转染Nrf2 siRNA质粒后,人参皂苷的治疗作用消失。结论人参皂苷通过促进Nrf2核转位,调控Nrf2/ARE信号通路发挥其抗波动性高糖所致内皮细胞损伤的氧化应激损伤作用。Objective To investigate the effect of ginseng on apoptosis of human umbilical vein endothelial cells (HUVECs) exposed to intermittent high glucose and the underlying mechanism. Methods HUVECs were eultured and divided into 5 groups: control group, intermittent high glucose group, and low(12.5μg/mL), middle(25μg/mL), high(50μg/mL) dosage of ginseng treatment group. The nucleoprotein of Nrf2 and its downstream antioxidant enzymes such as heme oxygenase-1(HO-1), γ-glntamyl cysteine synthetase(γ-GCS) and NAD(P)H quinone dehydrogenase I(NQO1) were detected by Western blot. The mRNA level of HO-1, γ-GCS and NQO1 were detected by RT- PCR. Western blot was used to detect the nucleoprotein of Nrf2, and HO-1, γ-GCS and NQOI after HUVECs transfected with Nrf2 siRNA. Results After the treatment of ginseng, the nucleoprotein of Nrf2 and the level of mRNA and protein of HO-1, γ-GCS and NQO1 increased(P〈0.05), hut the reatment effect of ginsenoside disap- peared after HUVECs transfected with Nrf2 siRNA. Conclusion Ginseng may have positive function of anti-oxida- tive stress on apoptosis of HUVECs exposed to intermittent high glucose by promoting nuclear translocation of Nrf2 and regulating Nrf2/ARE signal.
