摘 要:目的研究三生合欢饮抗抑郁作用及其对肠道菌群的影响。方法健康雄性昆明种小鼠60只,随机分为空白组、模型组、百忧解组及三生合欢饮高、中、低剂量组(15.04g/kg、7.52g/kg、3.76g/kg),每组10只。采用慢性应激和孤养的方法复制慢性应激抑郁小鼠模型。第21天测定小鼠体质量增加情况、糖水消耗、行为学以及脑内多巴胺递质含量等指标。第22天,采集各实验组小鼠粪便样品,采用ERIC-PCR技术获得反映肠道微生物群落结构特征的ERIC-PCR指纹图谱。结果与空白组比较,模型组小鼠在第14天后体质量增加显著降低[14天:(16.22±0.80)g比(20.34±1.06)g];21天:(20.04±1.62)g比(22.71±1.35)g;P<0.01),与模型组相比,三生合欢饮高剂量组[14天:(17.93±0.94)g,21天:(22.58±2.15)g]、百忧解组[14天:(17.16±0.93)g,21天:(20.97±2.89)g]小鼠体质量增加明显高于模型组(P<0.01);与空白组[14天:(3.94±0.80)m L,21天:(2.62±0.56)m L]比较,模型组小鼠糖水消耗[14天:(2.75±1.01)m L,21天:(2.05±0.76)m L]明显降低(P<0.01,P<0.05)。与模型组比较,第21天后三生合欢饮高﹑中﹑低剂量组小鼠糖水消耗量[(3.66±1.05)m L、(3.10±0.70)m L、(3.10±0.70)m L]显著增加(P<0.01),百忧解组小鼠糖水消耗[(3.34±1.84)m L]亦明显增加(P<0.01);与空白组[(248.67±43.64)次/s、(35.67±20.66)次/s]比较,模型组小鼠水平活动和垂直活动次数[(207.55±46.07)次/s]明显减少(P<0.05),游泳不动时间[(77.44±64.79)次/s]明显增加(P<0.05);与模型组比较,三生合欢饮低、中剂量组小鼠自主活动次数[(253.10±40.72)次/s、(251.90±45.39)次/s]显著增加(P<0.01),低剂量组小鼠游泳不动时间[(44.70±40.48)次/s]明显减少(P<0.05);与空白组[(1.21±0.34)μg/m L]比较,模型组多巴胺(DA)递质含量[(1.12±0.17)μg/m L]显著减少(P<0.05);与模型组比较,三生合欢饮低剂量组DA含量[(1.31±0.25)μg/m L]明显提高(P<0.05);EKIC-PCR指纹图谱显示,模型组与空白组在500bp以Objective To investigate the Antidepressant effect of Sansheng Hehuan decoction and its effect on intestinal microflora. Methods Sixty healthy male mice were randomly divided into 6 groups: control group, model group, fluoxetine group, Sansheng Hehuan decoction groups (high, medium and low dose). Chronic depression rat model was induced by chronic stress and isolated breeding. Weight gain, the autonomous activity, and monoamine neurotransmitter were determination on Day 21. Fecal samples wee collected in each experimental group of mice to obtain ERIC-PCR fingerprinting by ERIC-PCR techniques for analyzing intestinal microbial community structure. Results Compared with control group, model group rats' gained weight was significantly reduced after 14 days (16.22±0.80g vs 20.34±1.06g, P〈0.01) and 21 days (20.04±1.62g vs 22.71±1.35g, P〈0.01). Compared with model group, rats' gained weight in high dose Sansheng Hehuan decoction group was significantly higher on Day 14 and 21(Day 14: 17.93±0.94g; Day 21: 22.58±2.15g, P〈0.01 ), as well as in fluoxetine group(Day 14: 17.16±0.93g; Day 21: 20.97±2.89g, P〈0.01). Compared with control group, model group mice had significantly lower sugar water con- sumption on Day 14 and 21 (Day 14: 2.75±1.01mL vs 3.94±0.08mL, P〈0.01; Day 21: 2.05±0.76mL vs 2.62±0.56mL, P〈0.01); compared with model group, sugar water consumption of mice in high dose, middle dose, low dose Sansheng Hehuan decoction group mice increased signifieantly on Day 21 (3.66±1.05mL, 3.10±0.70mL, 3.10± 0.70mL, P〈0.01) and that in fluoxertine hydrochloride group also increased signifieantly(3.34±1.84, P〈0.05). Com- pared with eontrol group, the number of horizontal and vertical activities of model group miee decreased signifi- cantly(207.55±46.07/s vs 248.67±43.64/s, P〈0.05), but the motionless time in forced-swimming test of model group inereased(77.44±64.79/s vs 35.67±20.66/s); compared with model group, the number of autono
