摘 要:目的探讨右美托咪定对冠心病大鼠血小板功能的影响,评价其可能的心肌保护作用。方法 SD大鼠30只,随机分为正常对照组、模型对照组和右美托咪定组,各10只。高脂饮食和腹腔注射垂叶后体素建立冠心病模型。右美托咪定组大鼠尾静脉注射右美托咪定10μg/kg,连续5天,正常对照组和模型对照组给予等剂量的生理盐水。检测各组大鼠血浆血栓烷素B2(TXB2)和6-酮-前列腺素F1α(6-k-PGFlα)水平,评价右美托咪定对大鼠血小板功能的影响,分析肌酸激酶同工酶(CK-MB)和肌钙蛋白-Ⅰ(TroponinⅠ)水平,评价右美托咪定对大鼠心肌保护作用。结果与模型对照组比较,右美托咪定组血浆TXB2[(28.67±5.38)pg/m L比(38.25±7.79)pg/m L]、CK-MB[(10.31±1.81)IU/L比(15.40±2.94)IU/L]、TroponinⅠ[(10.07±0.97)ng/m L比(13.66±1.36)ng/m L]水平明显降低(P<0.01,P<0.05),6-k-PGFlα[(27.64±4.61)pg/m L比(21.19±7.72)pg/m L]明显升高(P<0.05)。结论右美托咪定可能通过改善TXA2和PGI2的不平衡状态,抑制血小板聚集,对冠心病导致的心肌细胞损伤起到保护作用。Objective To investigate the effect of dexmedetomidine on platelet function in rats with coronary heart disease and evaluate its possible role in myocardial protection. Methods Thirty SD rats were randomly as- signed to control group(C, n=10), model group(M, n=10), and dexmedetomidine group(Dex, n=10). High-fat diet feeding and intraperitoneal injection of pituitrin was performed on rats in model group and dexmedetomidine group to built coronary heart disease model. Rats in dexmedetomidine group were given intravenous injection of dexmedetomidine of 10μg/kg for 5 days, once a day. Control group and model group were given equivalent volume of normal saline. Serum TXB2 and 6-k-PGF1 alpha were detected to evaluate the effect of dexmedetomidine on platelet function, serum CK-MB and troponin I were measured to assess the effect of dexmedetomidine on myocar- dial injury. Results Compare to model group, the level of serum TXB2 (28.67±5.38pg/mL vs 38.25±7.79pg/mL), CK-MB (10.31±1.81IU/L vs 15.40±2.94IU/L), Troponin I (10.07±0.97ng/mL vs 13.66±1.36ng/mL) significantly de- creased(P〈0.01, P〈0.05), 6-k-PGF1 alpha increased(27.64±4.61pg/mL vs 21.19±7.72pg/mL, P〈0.05) in dexmedeto- midine group. Conclusion Dexmedetomidine can improve the unbalance of TXA2 and PGI2 and suppress platelet aggregation to attenuate myocardial injury in rats.
