摘 要:目的:建立反相高效液相色谱法测定斑蝥黄的含量。方法:采用日本岛津VP-ODS C18柱(150 mm×4.6 mm,5μm);流动相:甲醇-二氯甲烷=85-15;流速:1.0 m L/min;检测波长:450 nm;柱温:30℃。结果:斑蝥黄异构体能较好的分离,斑蝥黄在0.7μg^70μg/m L范围内呈现良好的线性关系(r=0.9998);重复性试验测得的精密度RSD=0.11%(n=5),平均回收率为99.6%(RSD=0.30%,n=9)。结论:该方法操作简便、灵敏、准确,能够满足检测斑蝥黄含量的要求。Objective: To establish an RP-HPLC method for determination of canthaxanthin content. Method: Japan Shimadzu VP-ODS C18 column (150 mm × 4.6 mm, 5 μm) was adopted with the mobile phase methanol : dichloromethane=85 : 15 at the flow rate of 1.0 mL/min; detection wavelength was at 450 nm, and the column temperature was 30 ℃. Results: The main peak was well separated from adjacent impurities and the linear range of the method was 0.7 lag-70 μg/mL (r=0.9998); RSD of repetitive test was 0.11% (n=5); the average recovery was 99.6 % (n=9). Conclusion: The established method was sensitive, accurate and specific, which can be used for canthaxanthin determination.
