摘 要:LIM结构域是在展示不同生物学作用的多种蛋白质中发现的进化保守的双锌指基序。LIM蛋白可以在细胞质和细胞核中介导蛋白质与蛋白质的相互作用。Ajuba属于LIM结构域蛋白家族第三类中的Zyxin蛋白家族,其参与细胞外基质的装配,并与相关蛋白一起调节连接细胞外基质和细胞骨架的靶基因蛋白表达。并介导广泛的生物学功能,包括调控细胞周期、细胞粘附与连接、细胞迁移、细胞增殖和凋亡以及细胞分化等。文章通过对LIM蛋白家族结构,分类及其功能的介绍,从而进一步对Ajuba蛋白的发现,结构及生物学功能在癌症和糖尿病的临床应用上进行综述。The LIM domain is an evolutionarily conserved double zinc-finger motif found in a variety of proteins with different biological functions. LIM proteins mainly mediate protein-protein interactions in the cytoplasm and nucleus. Ajuba is structurally a member of the third LIM family. It belongs to the family of Zyxin proteins which is involved in the assembly of the extracellular matrix. Together with assoealte proteins, Ajuba regulates expression of proteins that links the extracellular matrix with cytoskeleton. It also mediates a wide range of biological functions, including regulation of cell cycle, cell adhesion and attachment, cell migration, cell proliferation and apoptosis, and cell differentiation. In this paper, the discovery, structure and biological functions of Ajuba protein were reviewed in respect to its potential clinical applications in cancer and diabetes treatment.
