摘  要:色素上皮衍生因子(PEDF),属于丝氨酸蛋白酶抑制剂家族成员之一,广泛表达于机体各种的组织、器官,如视网膜、肾脏、肝脏、脂肪、肌肉、骨骼;与人体多种代谢性疾病,如糖尿病、肥胖症、代谢综合征、肝功能紊乱、炎症、肿瘤等相关。PEDF与不同受体结合发挥不同的生物学作用,包括抗血管生成、抗氧化应激、抗纤维化等。细胞表面存在两种主要的PEDF受体:PEDFR(由PNPLA2基因编码)和层粘连蛋白受体(LR)(由RPSA基因编码),PEDFR的磷脂酶A2活性与三酰甘油脂肪酶性分别介导PEDF的不同生物学功能,而LR主要介导PEDF的抗血管生成作用。在细胞、动物学研究中发现,PEDF对糖尿病微血管有保护性作用,可能参与早期糖尿病肾病的病理改变。Pigment epithelium-derived factor( PEDF), a member of the serine proteinase inhibitor (serpin) family, is widely expressed in tissues, organs, such as the retina, kidney, liver, fat, muscle, and bones ; PEDF is related with a variety of metabolic diseases such as diabetes, obesity, metabolic syndrome, liver dysfunction, inflammation, tumor and other diseases. Different PEDF receptors elicit divergent signals to cause different biological effects ,including anti-angiogenesis ,anti-oxidative stress,anti-fibrosis and so on. PEDFR( encoded by PNPLA2) and laminin receptor (LR) (encoded by RPSA) are two major receptors of PEDF on the cell membrane. LR mainly mediates the anti-angiogenic effect of PEDF, while the phosphohpase activity( PLA2 ) and triglyceride lipase activity(ATGL) of PEDFR mediate the different biological functions of PEDF respectively. In cell and animal studies,it's found that PEDF has a protective effect on diabetic microangiopathy and may be involved in the pathological changes of early diabetic nephropathy.
