摘  要:目的应用原位杂交技术观察当归补血汤对移植肌卫星细胞受体鼠造血功能重建的影响。方法对4组受体鼠的骨组织石蜡切片进行原位杂交,探究Y染色体的存在及阳性细胞表达率。结果除空白对照组,其余3组(移植肌卫星细胞+灌注生理盐水组、移植肌卫星细胞+灌注等倍当归补血汤组、移植肌卫星细胞+灌注5倍当归补血汤组)均呈阳性反应,且阳性细胞表达率差异具有统计学意义。结论原位杂交技术可以证明雌性小鼠骨组织中的Y染色体来自于雄性供体肌卫星细胞,当归补血汤干预有助于受体鼠造血功能重建,为后续研究提供进一步的实验室依据。Objective To study the effect of Chinese angelica blood tonic ( CABT) on the hematopoietic reconstruction of mus-cle satellite cell ( MSC) in allograft mouse by in situ hybridization. Methods Y chromosomes were detected by In situ hybridization in paraffin sections of bone tissue from four groups of receptor female mice. Results: Except for the control group, Y chromosomes were detected in the other three groups (MSC + normal saline, MSC + equivalent CABT, MCS + 5 times of CABT), and the difference between control and CABT groups is of statistical significance. Conclusion: The Y chromosomes in the bone tissue of the female mice were identified from the MSCs of male donors by in situ hybridization, which confirmed that CABT facilitated hematopoietic reconstruc-tion in the receptor mice. This provides a basis for further experimental study of CABT function.
