摘　　要：目的摸索提高微小组织冰冻快速制片质量的方法。方法针对小组织冰冻切片中常常出现组织切完、多粒组织切片不全、冰晶、染色不均匀等问题,采用预先制作冷冻平台、快速冷冻等方法来制作优质的冰冻切片。结果组织铺平可使组织切片完整,快速冷冻可减少冰晶形成,使组织结构更清晰,及时固定和加温染色可使细胞核染色更鲜艳,核、质对比明显。结论组织铺平、速冻、固定、染色等都是提高微小组织制片质量的关键。Objective To explore methods to improve the quality of quick frozen sections of small tissues. MethodsProblems occur frequently in the preparation of frozen sections for small pieces of tissue, such as tissue depletion, incomplete sectioning of multi-ple tissues, ice crystals and uneven staining. To make high quality frozen sections, several approaches were tried out including prepa-ration of the frozen platform in advance and quick freezing. ResultsFlattering tissue on frozen platform helps to keep tissue section in-tegrity; quick freezing helps to reduce ice crystals in tissue and produce nice sections; brighter cell nuclei and strong stain contrast be-tween the nucleus and cytoplasm were obtained by fixing tissue in time and warming up the staining solution. Conclusion Tissue flat-tening, quick freezing, fixation and staining are all critical to improving the quality of frozen sections of small tissues.
