唇香草挥发油急性毒性实验研究

摘　　要：目的探究唇香草挥发油的急性毒性。方法采用水蒸气蒸馏法提取唇香草挥发油,将90只小鼠随机分为9组,每组10只,给药组小鼠灌胃剂量分别为10 000、8 000、6 400、5 000、3 000、2 000、1 000、500mg/kg体质量唇香草挥发油溶液,空白组小鼠灌胃同等剂量的蒸馏水,按照0.2mL/10g体质量每天1次,连续灌胃7d,第1次给药前禁食不禁水12h,给药后密切观察死亡数1d,第2天以后每天观察2次,连续观察14d,记录各组小鼠灌胃前后体质量。通过小鼠死亡数量来计算LD_(100)、LD_(50)、LD_0。结果给药期结束,给药剂量为8 000mg/kg体质量组小鼠全部死亡,给药剂量1 000mg/kg体质量组小鼠全部存活,与空白组小鼠比较,各组小鼠体质量变化差异无统计学意义(P>0.05);经计算小鼠全数致死量LD_(100)=8 000mg/kg,小鼠0数致死量LD_0=1 000mg/kg,小鼠半数致死量LD_(50)=4 122.33mg/kg,置信系数a=0.05时的置信区间为:3 560.56<LD_(50)<4 772.73mg/kg。结论唇香草挥发油毒性低,临床安全剂量还需进一步探究。Objective To study the acute toxicity of the essential oil fromZizphora clinopodioides Lam.Methods Essential oils were extracted fromZizphora clinopodioides Lam.by steam distillation.90 mice were randomly divided into 9 groups,with each group 10 rats.The dosage of mice subjected to oral gavage was 10 000,8 000,6 400,5 000,3 000,2 000,1 000 and 500mg/kg,respectively.Blank control group was given the same amount of distilled water,according to the body mass of 0.2 mL/10 g,1 times per day,continuous oral gavage for 7days.First time before giving the drug,make the mouse fast but not water for 12 h.After the oral gavage,we observed the death toll every day,and recorded 2 times every day and continuously observed for 14 days.The body weight before and after oral gavage was recorded in each group.LD_（100）,LD_（50）,and LD_0 were calculated by the number of deaths in mice.Results At the end of administration period,the dosage of 8 000mg/kg group mice all died,and the dose of 1 000mg/kg mice survived,compared with the blank control group,there was no significant difference of body mass between the mice in each dose group（P〉 0.05）.LD_（100）=8 000mg/kg,0 lethal dose LD_0=1 000mg/kg,LD_（50）=4 122.330 0mg/kg,a=0.05 when the confidence coefficient confidence interval：3 560.559 9 〈LD50〈4 772.733 8mg/kg.Conclusion The essential oil of Zizphora clinopodioides Lam.was shown to be low toxic,and the clinical safety dose needs further investigation.
