摘  要:目的探讨新疆地区支气管哮喘(Bronchial Asthma,BA)患者白介素-4(IL-4)基因启动子区甲基化的差异。方法选取2014年1月-2016年10月在新疆医科大学第六附属医院呼吸科收治的BA患者80例为试验组,其中男性42例,女性38例,平均年龄(39.2±3.8)岁;选取同期门诊体检健康人群80例为对照组,其中男性44例,女性36例,平均年龄(40.1±2.6)岁;采用甲基化特异性聚合酶链反应(Methylation Specific PCR,MSP)检测IL-4基因甲基化(碱基C经亚硫酸氢盐修饰后变成U,但已甲基化的C则不再改变,据此设计两对不同的引物PCR扩增可检验其差异)。结果与对照组比较,试验组IL-4基因甲基化率较高,差异有统计学意义(P<0.01),试验组IL-4基因启动子区完全甲基化率为42.5%(34/80),未甲基化率为25.0%(20/80),甲基化和未甲基化共存率为32.5%(26/80);对照组IL-4基因启动子区完全甲基化率为11.3%(9/80),未甲基化率为46.3%(37/80);甲基化和未甲基化共存率为42.5%(34/80)。结论 IL-4基因甲基化程度在BA患者发病中有一定作用。Objective To investigate the methylation of interleukin-4(IL-4)gene promoter in patients with bronchial Asthma(BA)in Xinjiang.Methods 80 patients with BA were collected from January 2014 to October 2016 hospitalized in the s department of Respiratory of sixth Affiliated Hospital of Xinjiang Medical University.The average age was(39.2±3.8)years old,including 42 males and 38 females;as healthy control,80 subjects were selected.The average age was(40.1±2.6)years old,including 44 males and 36 females.Methylation specific polymerase chain reaction(MSP)was used to detect the methylation status of IL-4gene.Results Compared with the control group,experimental group,IL-4 gene methylation rate was shown to be significantly higher(P 0.01),the methylation rate of IL-4gene promoter in experimental group was 42.5%(34/80),non methylation rate was 25.0%(20/80), methylation and non methylation coexistence rate was 32.5%(26/80);Methylation rate of IL-4 gene promoter in control group was 11.3%(9/80),non methylation rate was 46.3%(37/80);methylation and non methylation coexistence rate was 42.5%(34/80).Conclusion The methylation level of IL-4 gene may play a role in the pathogenesis of BA.
